For the best combo, I would say yes. However, feel free to explore any variations that might work for you. The series is meant to provide tips to help with your own build process. I haven't fully explored it with other builds myself.
If you mean SST, either Slayer or Gladiator should work well. If you want to use the Transcendent Flesh and Timeless Jewel combo, go with Scion. For more tankiness, the new Gladiator is a good pick, and you can add Pathfinder for extra quality of life. It really depends on your playstyle and what's important to you
