I have eaten 25-50 gm of natto almost every day for 20 years. According to the USDA, this amounts to something like 5-10 mg of polyamines/day. Thanks for these videos.

Another great video, Michael! Where do you get spermidine in your diet? Do you source it from wheat germ, mushrooms, broccoli, soy, or something else? There is one study on spermidine in the human diet and its potential impact on aging - American Journal of Nutrition: "Higher spermidine intake is linked to lower mortality: a prospective population-based study" Authors: Stefan Kiechl, Raimund Pechlaner, Peter Willeit, Marlene Notdurfter. There is also this new study that has a lot of references published in 2021: New Insights into the Roles and Mechanisms of Spermidine in Aging and Age-Related Diseases by Yu-Qing Ni. Thanks Bob, almost exclusively mushrooms, but note that spermidine is also produced by the gut microbiome. I wanted to focus on circulating levels, not dietary intake (as you mentioned), as there is published evidence that dietary intake doesn't impact circulating levels: pubmed.ncbi.nlm.nih.gov/37111071/

@@conqueragingordietrying1797 Thanks Michael. Isn't this study about subjects taking spermidine pills and not getting it from whole foods? As you have pointed out, we are better off getting specific nutrients from whole foods as they are better absorbed and are more effective than when they are isolated in pill format. Bryan Johnson gets his spermidine from chlorella in his blueprint plan. Ever thought of adding chlorella to your food list or are you convinced dietary intake of foods with spermidine doesn't matter?

@@conqueragingordietrying1797 This paper also argues the dynamics of the pathway are more complex: supplementation doesn't increase spermidine levels directly, but spermidine may act as a prodrug which is converted to spermine and this goes into circulation and may enter the cell and be converted back to spermidine. It's possible there is a large reservoir to fill and yes you wont measure changes in serum, but other biomarkers might improve.

Some studies have shown an association (not causation) for spermidine levels on stroke risk, but conversely, some studies don't show that association: pubmed.ncbi.nlm.nih.gov/38101276/

Just as a heads up: "An overview of polyamine metabolism in pancreatic ductal adenocarcinoma" appears to suggest that high levels of spermidine (and other amines) may assist the growth pancreatic cancer...? I also found a study called "The effect of spermidine on autoimmunity and beta cell function in NOD mice" that seemed to show that mice on spermidine had higher incidences of diabetes. Obviously very specific type of mouse related but might be worth a look...?

Definitely, and thanks @bl3681. Spermidine is produced by the gut microbiome, so I'd bet that's contributing to the too-high is bad for health-story.

Also keep in mind the local environment in cancer can be very different to the circulating. In the NOD mice paper they saw spermidine increase in plasma 1.8-fold, p

@@bl3681 I don't believe anything like that exists because its hard to measure changes in serum. The epidemiological studies indicate (the one that Michael presented in the previous spermidine video) mid range is ~10mg/day, upper ~11.5mg/day as a start.

After doing some more digging, studies showing lifespan extension in mice use much lower concentrations in the drinking water around 0.3mM, or they use higher doses for short times. If water consumption is similar between the mice models it appears 10mM across the lifespan is a mega dose for NOD mice.

MIchael, I've just started using Iollo too but having a bit of trouble interpreting results. My spermidine results for example was fairly good, 4.8 uM and my Taurine was ~100 uM, which Iollo interpreted as optimal. But taurine in the literature and in typical blood tests panels (Quest and Labcorp) is for plasma (or serum) while we provided dried whole blood. Taurine values for whole blood in the literature is a lot higher than plasma for example. Iollo web site does not clarify nor does their AI. From what you know should I be interpreting Iollo results as serum or plasma levels as opposed to whole blood. Even the literature is a bit sloppy in using these terms, or it's my lack of understanding.

Hi Peter, I wouldn't compare iollo's concentrations to venipuncture-published results, instead focusing on avoiding age-related changes, i.e. decreases for taurine, increases for kynurenine/tryptophan, oxidative stress-related, or uremic metabolites. That's how I'm using their kit...

@@conqueragingordietrying1797 Thanks. But the second concentration graph (2019 paper) was based on plasma in units of nanomole/L (per Table 2) vs iollo result of micrornole/L. Will get back with Iollo as we need to be able to use their result with the literature.

The Joaquim et al 2019 cohort you compare to are average age 75, much older than yourself. Here is a new paper with a younger control cohort: Positive Correlation between Relative Concentration of Spermine to Spermidine in Whole Blood and Skeletal Muscle Mass Index... Sanayama et al Biomedicines 2023, 11(3), 746. They use controls of average age 45 and compare to dialysis patients. Interestingly it shows the controls have a higher Spermine to Spermidine ratio.

It was first discovered in semen. I think the person who made the discovery named both spermidine and spermine. The chemical name is N1-(3-Aminopropyl)butane-1,4-diamine.
