Walk through how to run RoboColumns experiments on the Synthace Digital Experiment Platform. These small scale column chromatography experiments that can be automated on Tecan liquid handling systems.
Synthace uses visual building blocks called elements you can use to describe different parts of our process, different materials that we're using. Link elements together to describe a workflow to characterize an entire experiential method. Each building block opens up and allows you to parametrize the different aspects of your experiment. It’s easy to make changes and run do complex methods that have multiple different parameters or multiple different factors in the same experimental run.
It helps you better leverage automation tools and transcribe it to a particular piece of automation in your lab. In this example we run a RoboColumns experiment on our Tecan Fluent. We select the device from our library, enter the configuration of our instrument, and allocate labware.
Synthace does all the hard work by transcribing your method into a set of executable automation instructions for your chosen instrument. You don’t have to write any scripts, and changes are as simple as a few clicks. Synthace then generates a simulation for you that you can then drive on a Tecan instrument.
Individual volume transfers are calculated for you and device specific liquid handling instructions are ready to drive your execution. The graphical preview of your experiment shows you everything you need to set up your run. Once you’re ready, simply schedule this for execution in the lab, or jump back into the method and make changes. Let’s say your Tecan Fluent is occupied today, and you want to run this exact same method on a different instrument - you can do so. Just select a different instrument. Synthace will transcribe the method for your new device.
Your results tab is going to show you all the data collected from your process. In RoboColumns purification experiments, collecting data points and associating those fractions with your samples and metadata is tedious and frustrating. Synthace does this process automatically by ingesting all of that data, automatically structuring and aligning it in a graphical user interface for you to analyze and inspect.
Finally you can review your chromatography results from your RoboColumns experiment. See chromatograms, heat maps for the collection plates that you’ve analyzed, and even data layered on top of those plates so that you can understand and inspect for particular samples in particular wells. All your data is pre-structured for you in an aligned data set where the output readings are associated with fractions collected and all the details about those fractions and how they got there. This saves you tons of time in data alignment and structuring.
