Can this work with 12 v relay? and how to calibrate the lowest temperature ?

yes it can,to calibrate the lowest tempreture,u need to turn the potentiometer unticlockwise and each stage u turn the potentiometer heat up the sensor ,it should take longer time to activate the relay

How to determine desired temperature?

the best way is to use voltmeter ,to set a desired premature

Hello sir. Are you using 12 V or 5 V fan?
