You didn't mention it, but a big reason Neurodegenerative diseases are on the rise is also because fewer people are dying from Cancer and Cardiovascular (heart attacks and strokes) along with many diseases managed long term like HIV and Diabetes. So basically, we've increased the odds that we'll find out if we get a neurodegenerative disease like Alzheimer's.

I'm wondering how much these treatments will cost... I often wonder how the creators of breakthrough medications in America can sleep at night, knowing that after they slap patents all over their invention, they proceed to inflate medication costs well above what it is worth, so they can profit off individuals in their own nation as well as medical services in other countries around the world.

When I was a kid, dying at 60 years of age or early 60s was pretty regular... wasn't a big shock. If you made it to 60 or so years, was sort of considered more or less a good go, that people had their innings so to speak. Today, if you don't make it to 80, it's sorta considered too soon and a shame. So it's not surprising that a lot of late age ailments are on the rise that didn't exist much before or in the same numbers.

I work in a Long Term Care Home in Canada, where like 60+ percent of our residents have some form of Dementia, so its pretty encouraging to hear that there are new drugs that could actually help some of these people. Its a really sad and lonely way to die

One lesser AI headline I liked this year was its use in the Herculaneum scrolls -- a massive collection of historical texts burnt to near ash BEFORE the centuries buried them. Yet, with modern scanners & AI trained to pick out the ink from the char, they've slowly started reading what this library had to say. [Just "purple" so far, but very early days...]

Fun fact Joe, when a drug ends with "mab" it's usually a Monoclonal AnitBody that targets a specific thing (like amyloid plaques in this case). Also it's usually pronounced mab like the a in nap, "ahh" ❤❤

I wish those medicines were out eight years ago. I was the hospice care for my grandmother and grandfather that both died from Alzheimer’s. It’s a very sad way to die.

I thought this was so exciting. I know a young girl with this terrible disease. Casgevy, a cell-based gene therapy, is approved for the treatment of sickle cell disease in patients 12 years of age and older with recurrent vaso-occlusive crises. Casgevy is the first FDA-approved therapy utilizing CRISPR/Cas9, a type of genome editing technology

That's interesting. Are they using mRNA lipid nanoparticles as vectors? They've also used mRNA vectors for CRISPR cas9 to cure live rats of HIV, though I can't seem to find the report anymore?

Both arms of the phase 3 trial for lecanemab saw participants worsen. The lecanemab arm worsened a little bit slower. It definitely cleared plaques--so it wasn't a question of successful "target engagement". I still think there is plenty of reason to question the amyloid theory of AD, basically.

I hope there are science teachers out there screening these for their students each yr. I wish that U📺 vids like this existed when I was a kid. So much of science is delivered as existing knowledge and as a kid it’s often difficult to appreciate how our understanding is changing in the here and now. It wasn’t until well into Hubble’s existence that I really started to appreciate how significant it’s contributions were. It would have made science so much more engaging to get an annual summary and idea of what some of the key areas that those discoveries are enabling further development on going forward. I’d like to think I would have listened and been inspired.

Joe, your fireplace opening is too large and you aren’t getting enough replacement air. Common builder mistake. The smoke stains on the stonework are the giveaway. Cheap fixes: Crack a window in the room with the fireplace. The house is going to pull replacement air from everywhere otherwise. Smoking when lighting your fireplace can many times be avoided by settling a hair dryer, etc., in a position where it’s blowing hot air up the flue for 5-10 minutes. To reduce the fireplace opening, you can add a set of glass doors.

My grandfather has Alzheimer's and he had a stroke and he got better with his memory like he had it for five less years or so. It coming back but the doctors think that either the blood cleared that plack stuff away or his brain making new pathways went around where the worst part of his brain where.

Now all we have to do is get doctors to bother diagnosing it within eight freakin years. At this point, it's common enough and disabling enough that screening should probably be mandatory, like colorblindness.

@jcortese3300 Yes. We also need women to understand that debilitating pain is not normal, and we don't have to just live with it! I suffered from the age of 13 to 34 without realizing that what I was going through was a problem. I had DIENDOMETRIOSIS my ovaries were adhered to my pelvis and bowels.... I never felt more validated than the day of my surgery, knowing it was all real.

I found the cracking / clicking noise to be extremely distracting and unpleasant. I had to bring up my audio equalizer and put a low-pass filter on to be able to finish watching it.

I take care of people with Alzheimer's. It's great that there could be a cure, but I think about the people who've already gone, who I didn't get a chance to know in their right minds. Who I wandered the halls with in the middle of the night, and who I've formed bonds with. Many of them mistake me for family or an old friend. I'm glad I can bring them comfort. But I wish I could've known them in their prime.

For whatever reason, I first heard the statistic at about 7:30 as, in the US nearly half of people with Alzheimer’s are also caregivers of people with Alzheimer’s. I guess this episode needed to be a little cozier for me today. Lololol
