Methylfolate side effects can occur more common in some people than others. In this article and video we discuss these side effect, questions that come up when using methylfolate and give some reason why they might occur. If you have an mthfr gene mutation you may have personal experience with these or heard about them. Methylfolate can be a very potent supplement for those with mthfr gene mutation but some people don’t seem to tolerate it as well as others.

The most common methylfolate side effects I see, as a functional medicine doctor, from methylfoalte can be broadly categorized as "over-methylation". Over-methylation put simply is the biochemical process that occurs from taking too much methylfolate or other methyl donors. Methylfolate is at the center of the methylation process and when optimized allows for all the positive effects of methylfolate.
However, too much mehylfolate will actually shut the methylation process off and lead to fatigue, muscle or joint aches, depression, etc. The same same symptoms that methylfolate supplementation typically helps with, can trigger those symptoms when given in too high a dose. This problem is very common in people taking deplin a very potent methylfoalte supplement. In fact, most people with mthfr gene mutations don’t need this much methylfolate. Some do but that would be an exception not the rule.
So we can place 3 common methylfoalte side effects into the overmethylation category and they are: Fatigue Joint and muscle aches Anxiety
Remember joint aches and fatigue can also be common symptoms of not enough methylfolate but when given too much the system flips leading to worsening of your symptoms. Most of the time this occurs when people are taking the methylfolate over a period of time. At first they feel better for a period of time and then all the sudden feel like it is not working or start having these symptoms. Anxiety can and does occur from over methylation too but some people can only tolerate small amounts of methylfoalte right from the beginning. In these cases you may not technically be in an over methylated state. This has to do more with their unique genetics and possible other things specific to their lifestyle and epigenetics. Another common methylfolate side effect is digestive system pain or discomfort when taking methylfolate.
Upon taking methylfolate or other b vitamins some people start to have abdominal pain or discomfort, headaches, rashes, or other symptoms. In these cases you may wrongly assume these symptoms are from the b vitamins themselves. While this could be the cause, a bigger consideration is to make sure you don't have a problem in your digestive tract. Small intestinal bacterial overgrowth, fungal overgrowth and other pathogenic bacteria can and do consume b vitamins and can trigger these and other symptoms. While digestive problems are one big cause for methylfolate side effects or poor response to MTHFR gene mutation treatment, there are many other health issues that can do this too. That is why we should emphasize and focus on bringing the entire person back into balance don't just focus on MTHFR alone. One methylfoalte side effect that many people often don’t consider is in response to histamine and allergies. When histamine is broken down in the body, the main enzyme that does this is called histamine n-methyltransferse or hnmt. When histamine is converted into methyl-histamine it becomes a more potent form of histamine before it is then deactivated and eliminated from the body. If the follow up stages of histamine clearance are not working people often have increased allergies and histamine symptoms like skin rashes, or increased allergies.

I have two abnormal mthfr gene mutations COMT CME CBS and others. My dietician suggested methyl folate 400 mg and SAMe 200mg yeaterday. I take 0.25 of Xanax prn for 9 months. 0-2 times a day. Which one should I try to take first. I am concerned about side effects and have to be able to work.

Yes... health issues can be a 1000 different things, and usually it is multiple things working together and affecting each other.... At least try to look at your wellbeing from all angles! Diet, excercise, stress relief, sleep, emotional wellbeing.... and that will be your best bet. It may take a while to feel better. Its been a long road here too, but I have def. learnt that focusing on your health from every direction is very important! Good luck

Wish I would have watched this video before taking DEPLIN! It made me go nuts and I had no idea it could be the cause. When I found out I had the double MTHFR mutation, all I read online was take methyl folate!!!! I have suffered from anxiety and depression my entire life and I was hoping I found an answer to feeling better!! So I started taking methyl folate and had my Dr prescribe DEPLIN. And it made me worse- I went nuts!. I took DEPLIN for 90 days before realizing I was still miserable and my anxiety was worse than before! So I started researching again and found out about over methylation! Now I take Folinic Acid and feel great.

You can also take plain ol' folic acid as long as you take riboflavin with it. That will convert it to usable forms. And by the way, folinic acid is being classified as a drug by the FDA, so very, very shortly you'll have to get it from your doctor.

Liked & subscribed! Taking a pill which contains Methylfolate and Methylcobalamin and it seems that I now have to drink more water than usual.

Thanks for the video. I've been researching a bit and found out a couple of sources that support the supplementation of methylfolate to be taken when there's b12 deficiency. That's what I'm unsure about, because that's exactly my case. Never tested for MTHFR mutation, so can't tell about it. But my blood tests show B12 deficiency whereas B9 values are within the reference range. Would you have any position on this point? Would there be any risks? Better to take B12 (methyl form) isolated or alongside methylfolate?

So i have fatigue and digestive issues (cramping, constipation, weight gain) and anxiety already. So am i meant to use methylfolate or folinic acid? Or is it a trial by error basis?

Hi I took orthomolecular methyl b complex and it changed my life. Only problem is I started getting extreme thirst for 6 hours, then next day joint pain, then muscle weakness for a couple days that mostly went away. I have long haul covid and lowered gut immunity but it has dramatically improved my health after one dose. I haven’t taken anymore just trying to figure out how to dose it. Thank you

Hi Jennie, you might want to do some genetic testing to see what your body is lacking. It sounds like it is was too much as well. This is just the type of side effect one would expect from too much methylfolate. Check out my video on MTHFR. These will probably be helpful.

I take meds for depression. I was depression free for 4 years. Last month my symptoms returned and i found out i am deficient in folate..I started folic acid 5mg daily. 3 days and i am great. I can feel meds working since they increase appitite. Then on 6th 7th day, i got worse and depressed. I have also new skin rashs. Do you think very high folic acid did that? I took 40,000mcg in 7 days. Like how much time does it need to settle a bit and get less in my body

This is very interesting! MTHFR homozygous here. Bad methylation... very clear issues and I actually have all these “side effects” without being on methylfolate, so clearly dealing with methylation issues! My homocysteine is now 43.6, so I am starting L-methylfolate today! I hope I see improvement and not anything worse than now..... I was wondering though.... can it be dangerous to take this if you are unsure about B12?

Spot on for me. Always around day 3 of 400mcg all the side effects kick in. All the symptoms. I don't have insurance and trying to figure all these things out on my own with the help of doctors like you who explain it in a way I understand, so thank you. I know it's not as ideal as having a specialist, but it's all I got. I'm going to try Niacin 50 mg to see if it helps, and by not taking it daily until I can tolerate it. On day 1 and 2, I feel particularly great. Too great by day 2. Day 3, the fatigue sets in. Night 3... wake up with strange muscle sensations, severe anxiety and extreme wakefulness and creativity, only muscles hurt to much to be productive. Day 4, eyes wake up severely dry and sinus allergies are back with a vengeance. I'm trying to understand all this information, but it can be overwhelming. It seems to reduce the severe headaches with I take methylcobalamin, or glutathione, but it's hit or miss.

Hi E! Thanks for your comments, I am happy to try to help. You probably already know but not everyone needs methylfolate. If you are finding it helpful early on maybe you just need to take it less often. For instance, you can try dosing it every 3 days instead of daily. That is an easy way to approach it. The more difficult approach would be to look at your other SNPs, health tendencies, and health problems to find out why you are so sensitive and what to do about it. The reason it is hard to give you specifics is everyone is different and you have to look at the whole picture not just MTHFR. Look into toxin burden - amalgam fillings? Flame retardants in food/clothing? Fluoridated water? Hg - F - Br - Ch wreak havoc on methylation pathways.

I took methylfolate 400 mcg for 3 days. I quit because while I slowed down and felt more calm, my ocd worsened. My hypothesis is that methylfolate decreased my catecholamine and serotonin levels.

Hi There! Thx for the video. I have tested yes for MTHFR (1 parent/1 gene only). I just started on 5MTHF from Seeking Health. It also has B12, B6 and others in it. I decided to minimize side effects by taking 1 capsule every other day. To get my body used to it :) Do you have any feedback on this method, and for how long? I was thinking 2 weeks... Thanks!

My doctor just put me on methyl factors, among other things, and I have been taking it for a week and a half and the last three days I've been feeling really fatigued. I let him know. Just waiting for a reponse. 😞

I'm doing methyl b12 injections and taking methylfolate. Trying to find right doses. Something is causing bad bloating. I've been told that's not enough methylfolate.. I've tried higher doses like 6 mg. That doesn't help. I don't know my gene situation. But if we need B12, what are we supposed to do with a bad reaction? Will this eventually get better on its own? Do you think bloating would mean I need more methylfolate?

My blood test showed I was deficienct in folate, B12 in mid-normal range. I try to eat a lot of vegetables but still deficient, so I will have to supplement. I know I am MTHFR heterozygous, so I was thinking straight up folic acid was probably a bad idea... Started taking methylfolate 800mcg daily, no problems, but would it perhaps be better to switch to folinic acid instead? Could I go higher with folinic acid and restore my levels to a safe level? Are side effects not a thing with folinic acid?

Its not as common with folinic acid but if you tolerate the methylfoalte, it's probably better.

I recently switched to a prenatal vitamin that has methylated folate from broccoli (my last one had folate). It's only been a few days but I have been having headaches, nausea, and just an overall unwell feeling. Should I just go back to taking folate?

Hi Ketra If it continues to happen yes you might want to stop it. However, if you are planning on getting pregnant and you have MTHFR you also should be taking methylfolate. In that case you want to find out why it is giving you headaches. I think it is a little odd that the methylfolate is said to come from broccoli. Maybe try a different one or methylfolate by itself. Generally for prenatal it would be 800 mcg. Hope that helps.

Follow up: the seeking health supplement has 1360mcg(DFE)800mcg of folate in it. Also - do the side effects often go away after initial intake? And is that a few weeks generally? I was between buying this supplement and Quicksilver Scientific Methyl Charge - which seemed to be "more natural"- using TMG betaine. I think instead of folate. I guess if the seeking helath doesn't work well for me, I can switch to quicksilver.

Hi KC! these are good questions. If you are having side effects or problems from the methylfolate they likely will not go away without doing something more deliberate. There is a lot of individual variation, however. Lower doses to start is better or other methyldonors. Hey, sorry for the late reply. Yes, I have gotten better. Not planning to take methyfolate again haha. A lot of my side effects are basically gone from the methyfolate. After doing some more research, I believe I was quite vitamin D deficient and started supplementing and health is improving more. @@catarunverma7525

If you take too much methyfolate and it shuts down the methylation pathway, is it because of methyl trapping, where you do not have enough vitamin B12?

Yes, not enough b12 to work with the methylfolate is one way people will experience side effects. Although it is not always clear if these side effects are due to excess methyl groups or down regulation of methionine synthase (methylation). The more direct way to down regulate the same enzyme is through increased SAMe compared to SAH. When this ratio increases the methionine synthase activity decreases. This is why blood testing can be helpful to differentiate the different scenarios. Does that make sense?

Thanks for the interesting video. I’m interested to find out how to get the test for the methylation mutation and what to ask for. I live in the UK. A couple of years ago I started taking 2g a day of trimethylglycine on recommendation of a nutritionist who thought I might have a methylation issue. After smelly urine for a few days that then became normal again my fuzzy head and constant tiredness much improved but is not as good as I think it could be. Should I try methyfolate and B12? I did find 5g a day of TMG was to much and made me feel weird. I now take 1/4 teaspoon (about 2g) and it’s great. I also have problems digesting fats. Is that related? Glutamine supplementation helps me a lot with digestion problems and food intolerance.

Hello Christopher, feeling better from the TMG suggests your body was deficient in choline, folate or other methyl donors. TMG is a methyl donors and it is a good practice to rotate them in your supplementation. This ensures you create deficiencies elsewhere. TMG is related to fat breakdown in that is can be use to make choline which is needed for bile acid production.

Now I’m really steady. I’ve just noticed Folic acid is added to so many foods. I might have to bake my own bread to avoid it and cut out cereals completely. Thanks again. Looks like the wrong type of B12 is even in my oat milk.

Hi, is it possible that l-melthylfolate can start working the the following day after taking it for the first time? I told my doctor it worked for me immediately but only lasted a week then I felted so down and depressed. Kind of like a crashed. She said it doesn’t work that fast, it takes months and that was a placebo effect. But I felt so normal and great. I started off with 15mg.

@@swintegrative I understand you can’t give medical advice on here. Since I have the defect MTHFR and BDNF genes, I plan on trying an anti-inflammatory diet (gluten free or Mediterranean diet) and either drop down to 7.5MG of MethylFolate or double to 30MG to see if that helps me. I’ll start of with the low dose and see if the new diet does anything. My current diet which I had since a teenager is extremely bad (fast food). It’s hard eating healthy when you travel for work. I m 16 year old teen and my mom's 2 months pregnant so, she was advised to take this medicine by the physician and i was sick so, i ate this iron & folic acid tablet of 500mg by mistake thinking it's the fever pill...i m so worried now, i remember my whole body was itching the night i had this in the afternoon but not many other symptoms are seen day 3 of this mistake but i m worried of any internal issue with my body...😫😥😥😥pls give an advice?

Yep, sounds like the folate is overstimulating your system and you are sensitive to it. Things like magnesium and non-stimulating b vitamins may be helpful. I would also consider a smaller dose. Try to find the minimum amount you can take w/o this occurring while still getting some benefit. Sometimes when you initially start methylfolate your body is just really sensitive and may take some time to fully utilize it. It's hard to say without a full picture of your genetics and health. I will do that. As for potassium it will not help fatigue (typically) and can be harmful in larger amounts.

Hi Dr, I have the mutation and my Dr recommended to take methyl folate but the one I am taking says folate 1,333 MCG DFE ( metafolin 800 MCG) so I don’t know if I am taking the right thing, can you please advise.

Hi Claret, Yes metafolin is the right type of L-methylfolate. Metafolin is like a a brand of L-methylfolate.

I was prescribed this due to neurapathy obtained from chemo. I'm very fatigued and have muscle aches, will it go away?

Hi Diane, it is very hard to say without knowing the full picture. If those came on after starting folate, it probably will not go away.

Maybe I haven't found the right supplement, but B12 (all forms) and Methylfolate supplements do fuck all for me. Surprisingly, folinic acid had a pretty cool effect on me that lasted only a day or two. Clarity, word recall was on point, immune system seemed to be decent, then bam gone. Getting tired of trying to figure this all out tbh.
