0:55 'Red' 5 volt and 'Black' ground. These two are input , whereas 'Yellow' signal is output? Output signal is how much in voltage?

It's depends of the viscosity of honey,the viscosity should be such that by flowing from input to output it can cause the rotation of turbine.

what will happen if water flows in opposite direction or will the output signal me same ?

The output will be not accurated if you change the water flows direction,the information regarding that is also return on datasheet of the sensor.
