So beautiful explained.All doubt cleared about yeast two hybrid system. Y2H system is used to study protein - protein interaction. Suppose we need to study about the interaction between Protein x and Protein y, then the 1st step is to ligate these two protein in two separate plasmid with Gal 4 AD and Gal 4 BD(Gal 4 AD and Gal 4BD are transcription factors which forms a heterodimer and bind in the promter and switch on the transcription and a reporter protein is formed which can either be a fluorescence gene or Lacz gene). 2nd step, Both the plasmids are transformed inside the yeast cell.(Note: Gal 4 AD & Gal 4 BD should be knockout from the yeast genome) Now, the yeast will form Gal4 AD & Gal 4 BD along with Protein x & Protein y in such a way that, only when the gal 4 AD & gal 4 BD will come closer to form a complex and bind in the promoter region when the two proteins Protein x & Protein y interact and form a complex. So, if the two proteins interect the reporter gene will give rise to reporter protein,so we will conclude that the Protein x & Potein y interact with each other.
