That's a great explanation thanks. I'm going to test using my sous vide today, along with an electric air heater to warm up my sab. I have tried letting my plates vent off condensation before putting the lid on, inside my sab, they were visibly very clean but did get some contam. Not 100% sue that was the cause but I'm cautious aout doing this..

This happen to me plenty of times, I wish I could able to share pictures of how condensation really effects bacteria on a Agar plates. You should wait when the agar haas cool down to 60-50 c. Then continue to pouring it to the agar plates. Another way is to let the lid open and wait for the steam to disappear. Do this right after the agar temperature has drop down to the desired temperature.

So, my situation is that I watch this video late, lol. I just poured 20 plates a couple of days already. How can I fix the condensation for the moment?..NEW SUB!!
@mushroomcult
@mushroomcult 4 года назад
You can dump the water out when you use the plates. If you have a flow hood you can let them dry in the air flow. Also store the plates up side down.

Can you just put a well colonized grain of rice on an agar plate is that ok? Or should I try to just get a bit of mycelium off of it instead? I’ve got 8 rice bags almost fully colonized, and 10 agar plates I want to keep this strain going and looking for the best way.
@mushroomcult
@mushroomcult Год назад
Great idea. This is a great way to save a cultivar and to quality control your grain spawn. Make sure you use aseptic technique to transfer the piece of rice

What about pre warmed/heated plates or jars and adding agar via a self sealing port?
@mushroomcult
@mushroomcult Год назад
Warming the plates is a great idea. That will reduce condensation to some degree. I'm not sure that adding the auger through an injection Port will decrease condensation. It could increase risk of contamination depending on your technique

HELP! So my plates are under a lamp for incubation and there is a layer of condensation on the lid. I've emptied out the water, but it's still condensing.
@mushroomcult
@mushroomcult 3 года назад
the issue is probably because the lamp is applying heat to the plates. most of the time there is no need to increase the temperature of the plates for incubation. normal room temperatures are perfectly fine. between 60f and 75f are perfectly fine.

@FreshfromtheFarmFungi 4 года назад
awesome work! Love the idea - is there a maximum limit to the volume of the water heater? Also, I recommend getting a laser thermometer that way you can see the temperature decrease in the agar without contaminating it - MUSHLOVE
@mushroomcult
@mushroomcult 4 года назад
I will plan on getting a laser thermometer so I don't need to guess any more. Im sure there is a maximum water volume but it is not in the produce instructions i may run a trial to find the max. the container i used in the video preheats to 124°f in minutes so i bet it can handle a pretty large volume.

Quick tip about sous vides, they are usually meant for 4-12 hour runs. It will get to exact temp 100%. But it takes a long time. Might be best to take from pressure cooker hot and put in sous vide to come DOWN to temp rather than up. obviously an ice bath would make this faster back n forth.
@mushroomcult
@mushroomcult Год назад
Exactly. The sous vide a is only for bring the temp down after sterilizing. It can hold the temperature longer if needed
