I took the third sample because my first urine was at 3 am, the second was at 5 am, and the third was at 8 am, and I started with it. Does this affect the validity of the analysis?

I have started taking Empagliflozin since Dec-2021. Before I started, back in that time my urine albumin / creatinine ratio was 7.23. But after taking Empagliflozin for 4 months, by April-2022; urine albumin / creatinine ratio has increased to 40.
