It's her most important flaw, and one she's almost put a finger on ("I simply do not understand how people don't see the world the way I do!"). But she hasn't really connected the dots, even if you can see in this episode she's starting to realize it a bit. She needs to mature, and burst her very sheltered bubble. Hopefully we'll get there ;-) Great reaction, as always! Don't be too hard on yourself, it's not always easy to pick up on everything when you're not familiar with the books and the characters inner thoughts. You're not the only one to think that Colin didn't have much of a romantic interest in Penelope before this and you're right. I'll give it to you as short and sweet as I can lol. They'd been friends from day one despite her hidden feelings for him and those feelings deepened as they matured into adults. After travelling to "find himself" and having empty sexual experiences he questioned the act of having sex altogether because he feels nothing during and after each one but keeps trying and plowing away. It's not until he kisses Penelope that he realizes he's not sexually fulfilled by anyone else because he had no emotional connection to them. Long story short, Colin is a demisexual and only capable of true sexual fulfilment with a partner whom he shares real feelings. Because he already cared so much for Penelope, the attraction clicked as soon as he kissed her. He's always had feelings for her, but unable to identify them due to not fully understanding his own sexuality.
