Hi, my name is Dr. Spin and I will try together with Dr. Scan to introduce a new technique to characterize and quantitate gene therapy products like viral vectors. This new technique is called density gradient equilibrium analytical ultra centrifugation or DGE-AUC. Dr. Scan, what is this new technique?

Density gradient ultracentrifugation, or DGUC is a tried and tested method to purify viral vectors and other biologics of very high purity. The same principles involved in purifying a sample by DGUC can be applied to characterize it.

Separation and purification are frequently the same thing. Consider a preparation of viral vectors with empty and fully loaded capsids. Your objective is to purify the therapeutically active product, which is the full particle containing the gene of interest. It can alternatively be thought of as separation of fully loaded and therapeutically active capsids from empty and inactive capsids. Either way, the process is based on the fact that fully loaded capsids have a higher density than empty capsids due to the inclusion of genomic material within the protein shell.

Yes, you can use the DGUC technique for viral vector purification in your ultracentrifuge. And then you can use that same technique to characterize your sample in the analytical ultracentrifuge or AUC. One technique for both steps.

How does that work?

When you spin a mixture containing particles of different densities such as empty and full capsids, and a solution containing a density gradient forming material like cesium chloride, a density gradient is formed. At the end of the experiment, the particles are separated in space with the high density particles lower down in the tube than the low density particles. The Optima AUC allows you to track the movement of these two species from start to finish.
