Antibody-Drug Conjugates (ADCs) are complex structures designed for targeted cancer therapy. They consist of three main components: a monoclonal antibody (mAb), a linker, and a cytotoxic payload. The monoclonal antibody selectively binds to specific proteins expressed on the surface of cancer cells, determining the ADC's specificity. A stable linker connects the antibody to a potent cytotoxic drug, ensuring stability in the bloodstream but allowing cleavage upon reaching the cancer cell. This cleavage releases the cytotoxic payload, disrupting cellular processes such as DNA replication or cell division, ultimately leading to the death of the cancer cell. Additionally, the Drug to Antibody Ratio (DAR) is crucial for understanding the composition and behavior of ADCs, as it directly influences therapeutic efficacy and safety. DAR determination involves enrichment and mass spectrometry analysis.
