@@OldSchoolChemistry, I thank you for your effort in making a contribution to knowledge that transforms humanity. As part of my PhD project, I was looking to identify an ester in some feed compound. I learned that it is possible to identify the presence of ester in organic samples through Ferric Hydroxamate Test. This test only shows the presence/absence of ester in your sample. It does not tell you the amount (concentration). That's where I came to know your channel. What if I measure the absorbance of my samples using a spectrophotometer and calculate the concentration of the available esters in my sample?

in my work it asks for Wavelength and Absorbance can you explain what each means and how they connect with each other? Thank you

Hi p h, wavelength is the distance between of a crest and crest of one wave of light. Wavelength is specific to the color of your solution. Absorbance, in essence, is the amount of light absorbed by the solution. The spectrophotometer is set to a wavelength to detect the amount of light absorbed by a sample at that specific wavelength. Once I have determined the conc. of the unknown sample using the curve, do I need to multiply it by the dilution factor ?

Hi Su zeO, no. This is because the concentration on the x axis was calculated considering the dilution factor. The concentration on the x axis should accurately represent the actual concentration put into the spectrophotometer machine. I hope that helps. Thanks!

Is that posible to record absorbance data greater than 1 during OD reading in spectrophotometer? because lambert beer law describes that absorbance greater than 1 can not give you straight line.

The absorbance is read on the spectrophotometer. It comes from the wavelength of the light absorbed which the machine reads as transmittance, and then is calculated to absorbance. Maybe I misunderstood your question. You may want to ask your question to your professor so they can clarify. Thanks!

Hi Mohamed, This is the only way with spectrophotometry. Remember once you have the plot you can simply use two points in the line to calculate concentration.
