Thanks so much for the comprehensive update. Mammal to mammal would include my fur baby species, the Canine. In North Texas we have been slammed with Canine influenza (H3N2, H3N8) since November 2022 and it's still going strong. I've received multiple notices about high risk areas such as groomers, vet clinics, dog parks and day care centers, the typical places that dogs visit. I would imagine that all eyes are watching this one. Hope it goes away and soon.
I’d be interested in information around transmission. The human cases, as I recall, are mostly poultry farm workers. The minks, and perhaps the seals (if it spread in a colony), were in close conditions with dense populations. That seems to indicate the probability of mammalian spread, but perhaps a higher infectious dose than other influenza viruses that have adapted to mammals or humans. (As a side note, the (caged) mink farm strongly suggests aerosol spread, barring something like aggressive spray down cage cleaning.) I wonder if anyone has studied infectious dose in susceptible birds/mammals? Also, if that is potentially relevant, whether there is knowledge regarding the likelihood of mutations that lower infectious dose (as appears to have happened relatively consistently in COVID, at least according to most reports). One might think there is at least a high-level tendency for that to happen, all other things being equal (which of course they aren’t : )
Why do you say it is uncommon? Please don't get the conspiracy theorists started about it being created in a lab for some bizarre purpose. Wikipedia notes, "The most widely quoted date for the beginning of recorded history of avian influenza (initially known as fowl plague) was in 1878 when it was differentiated from other diseases that caused high mortality rates in birds." That is before even the discovery of the structure of DNA.

In 2013 they performed gain of function on H5N1. They have a discussion on this at the NIH. It was stopped 4 years ago and there isn't much information after that.
Thank you for explaining the history of H5N1. It’s interesting to learn it started with a domestic group of birds. With all the sub-variants we have seen with SARS-2, it is easier to understand how H5N1 has evolved over time.

Do we know whether the mink farm employees wore PPE or at least masks when they did the culling? I would think they would have been careful not to be infected. In vitro testing of this variant on its ability to infect on human cells or humanized mice would be telling.

I'm not a virologist so this is out of my knowledge base but when looking at C-19 I did wonder if the spike protein is a more advanced form of HN ( Hemagglutinin-neuraminidase) as found in human influeza virus.
