Conditional gene expression using the Cre Lox FLEx vector switch!

The FLEx vector or "flip excision" switch uses the Cre Lox system to control gene expression. In this video animation, we'll review the basics of the Cre Lox system and introduce how the FLEx vector inducible gene expression system works so you can apply it in your own gene manipulation project.
In the Cre Lox system, a sequence of DNA can be excised, inverted, or translocated when it is flanked by a pair of loxP sites while in the presence of Cre recombinase. The orientation of the pair of loxP sites will determine the outcome of the gene recombination.
A FLEx switch has the gene or sequence of interest flanked by two different pairs of lox sites pointed in opposing orientations. When Cre is present, the sequence of interest will be inverted (using one pair of lox sites), with the intervening sequence (and the other pair of lox sites) excised irreversibly to prevent further Cre Lox recombination from occurring. This enables researchers to irreversibly turn gene expression ON or OFF when Cre is present.
A system where the gene of interest is in the inverted orientation (with gene expression initially OFF) enables addition of Cre to reorient the gene into it's correct orientation, turning expression ON. Such a system is called a Cre-ON system or DIO switch (Double-floxed Inverse Orientation). A system where the gene of interest is in the correct orientation (with gene expression initially ON) enables addition of Cre to reorient the gene into it's inverted orientation, turning a gene OFF. Such a system is called a Cre-OFF system or a DO switch (Double-floxed Orientation).
This video will also demonstrate how the Cre Lox FLEx vector switch can be used for tissue-specific gene expression, inducible gene expression, controlling multiple genes simultaneously and performing gene knock-ins.

Nicely explained!! I have a question..how to incorporate cre sequence in a mouse at the gene specific promoter? And also the lox p sites at the specific position?

How is it that these lox sites flank genes of interest without including other genes? It seems unlikely that so many genes are flanked by palindromic sequences, but am I incorrect in thinking that?

These lox sites are typically inserted into expression vectors and genomes by researchers in very precise locations. They do not exist naturally as flanking sequences of particular genes. Researchers will determine where they want the inversions to occur, and will either clone in the lox sites into their plasmids, or perform knock-in experiments to insert the lox sequences in the desired positions in the genome before carrying out their Cre-induced inversions. The gene must be inverted in the OFF (no expression) condition for the FLEx system. The Cre-Lox FLEx system controls expression by changing the orientation of the gene in relation to its intended promoter. For example, in the OFF position, the promoter would be in the forward direction while the gene (flanked by Lox sites) would be in the reverse direction, which would not lead to expression of the functional gene. After Cre is introduced to turn the system ON, the gene would be flipped into the forward direction and be able to be expressed under the control of the promoter. Hope this helps!
