Counting yeast cells with a hemocytometer
We will show you how to use a hemocytometer to count cells and share some tricks so that you can do it right. Various types of Hemocytometers exist. The most popular ones are Neubauer or Thoma Chambers (Neubauer improved chambers and Neubauer chambers).
For the exercise you will need the microscope, hemocytometer, its cover slip, ethanol, your sample and the pipette with tips.

hi. i have a doubt. for budding yeast, should i count the mother cell and daughter cell as two (2 budding yeast) if the daughter cell is same size as mother cell? and for smaller size daughter cell, the budding yeast counted as one (1 budding yeast)?

There are several scientific opinions on when to count a daughter cell as an individual cell. However, The American Society of Brewing Chemists (ASBC) recognizes budding cells as two distinct cells once the daughter cell reaches 50% or more in the volumetric size of the mother cell. Any less than this and the mother/daughter is counted as one cell. So the general rule is: Daughter cells are only counted if their area is 50% or more of that of the mother cell. If you’re doing the count manually, you will have to eyeball the size and make a judgment call.
