"Daddy, I cant unlock the door..." This is what my daughter said as she was trying to exit my 2008 Toyota Tundra this week. I simply responded, "just press the lock button over and you can open the door." As I listened to her toggling the switch back and forth, I decided I would investigate. I used the power door lock controls from the driver's seat to unlock all the doors. When I opened the back door, as soon as I touched the manual lock, the locking mechanism easily rocked back and forth. Very odd. I could use the power door locks to secure the door, but could not manually unlock it from the inside (which would cause a safety hazard in the event someone in the back needed to quickly exit the vehicle). From here, I figured the cable that runs to the back of this switch must have come undone. I removed the door panel to find that the dock lock assembly actually broke (which it's most likely inevitable given that it's plastic).
I picked up a new door lock assembly (Part Number: 69206-0C040 - Interior Door Handled (Left, Front, Rear) which is the assembly for the door lock and handle--you must purchase the entire sub-assembly as you can not purchase just the door lock. *This part runs approx. $30-$45.00 USD.

To change the door lock, start by removing the door panels. You will find a door trim panel upper clip near the base of the window and the door's edge (left side, if you're on the driver's side). Press the center pin to release the clip. Next, remove the dark plastic panel behind the door handle and remove the phillips head screw here. In the handrail, remove the small carpet panel to find one more phillips head screw and remove. Next, pull straight up on the entire plastic base that the automatic window switch sits in. *This step isn't required, but it's easier to disconnect the wiring harness for the switch at this time. Next, pull the top panel from the door (there are three clips holding this piece of panel onto the door). Once removed, you can disconnect the bottom panel. Behind this panel are nine (9) clips around the base of the panel--simply pull the panel away from the door. Now, the door panel is hanging near the base of the window. You can simply pull straight up and out to remove the panel. ***Do not pull too far from the door, you will need to disconnect the two cables running to the door lock and the door handle and a couple of wiring harnesses. The window switch harness is already disconnected if you did it previously (above) and last there is one more harness at the bottom if your door is equipped with a door panel light. *There is no wiring to the door speaker that connects to the panel. Flip the door panel around to access the door lock/handle sub assembly. You will find two phillip head screws to be removed. Next, there is a small tab at the top of the assembly that you press down to remove the assembly. Swap the sub-assembly, reconnect the wiring harnesses and make sure the cables (white is to the door lock, green is to the handle) are seated in the assembly and their housing, then reinstall the door panels.
