I have a question we usually save the data to the excel file but for the backup we also save the data immediately from gen5 ( raw data directly from the instrument). If someone forgot to do this is it possible to find a plate in the history? If yes ? How? Thank you so much in advance!

Hello Sr. I've already have the software in another computer a bit far from the instrument. I created the protocol in the instrument, but I was wondering if there's any possibility to export as an excel document and try to use it in the computer? Can we use the software for analyse the data that is already read by the instrument. Thanks for everything!!

Hi Rita Corrales For analyzing the data in other computer or laptop with Gen5 software you have to save your experiment data in .xpt format during data acquisition. you can't do this from excel format file. you will not be able to import your excel format (.exl) experiment file in Gen5 software to further analyze your data. sorry for this

Hi there! It sounds like you're having trouble finding the option plate layout in your Gen5 software. Don't worry; it's there, and I can help you find it. In your Gen5 software, when you open any protocol, you should definitely see four options: Procedure, Plate Layout, Data Reduction, and Report/Export Builders. If you can't see these options, it's possible that they are hidden. Here's what you can do: Look for a plus (+) sign button or an expand/collapse button in your software interface. Click on it to expand the options. This should reveal the Plate Layout option. Another thing to check is whether you have selected a specific task during experiment setup. Depending on whether you're setting up a standard curve, working with samples, or something else, the available options may change. Make sure you have selected an appropriate task to access the Plate Layout option. For more detailed guidance, I recommend checking out my other tutorial videos where I explain these software features in greater detail. Additionally, please consider watching my updated tutorial for this specific video, as it may provide further insights. I hope this helps you find the Plate Layout option in your Gen5 software! Sir, what what is corrected blank. Do i need to analyze corrected blank readings or should i go with blank only? The average is coming negative for all the samples after corrected blank.

Pathlength correction is used for Normalization of Absorbance when we use Cuvette or Biocell for detection of DNA or RNA. In this case many factors (like Liquid volume, Plate type and manufacturer, Buffer composition, Nucleic acid concentration, Temperature of the solution) affect the the absorbance . I suggest you please dont use pathlength correction use BLANK substraction only. In case of background correction using Primary (For Exm. 450nm) and Reference (For Exm. 650nm) wavelength. use data reduction option in Protocol and here in Delta transformation use Data In1: Primary (For Exm. 450nm) Data In 2: Reference (For Exm. 650nm) It substract OD value of Reference from Primary. Hello Sabir ji First open the device manager in your laptop or desktop and click on the down arrow sign \/ at Port (COM & LPT) and you can see COM port like COM2 COM4 or etc now connect the usb cable of Reader Machine and you can see new COM port like USB Serial Port (COM9). Now open the Gen 5 software and click on System and then open Instrument Configuration. In the Intrument configuration click on the View/Modify option. In the reader setting select the COM port from drop down list and choose as you have seen in the device manager that was port COM9 (as per your system) and click on ok . I hope this may help you.
