so replace the V (dominant) with a flatted II chord? does it work if you are not in a 2-5-1 structure?

It does, kind of. In this instance, Dbmaj7 going down to Cmaj7 does occur in lots of songs (see Girl from Ipanema) Also, chord qualities can be mixed and matched. See songs like Lady Bird (Tad Dameron) last two bars. And lastly, Bemsha Swing (T. Monk) also has lots of similar movements :) A good resourceS: www.jazzguitarlessons.net/blog/chord-substitutions

Can tritone be used on minor and major chords too

so....a melodic figure could include the Db dominant; the perception would be that of a V chord with alterations, proceeding to the Tonic; and a simple arpeggio would get the job done....
