You have to read research papers. Im not that experienced to suggest you. You need to talk with your research supervisor or teachers from your institute/lab.

Grow the seeds either in-vitro or in pots using leaf or internode develop shoots or develop callus from that develop shoots use the in-vitro leaf now to induce hairy root culture. Read literatures for specific conditions like hormones required etc

Yes. You can order them from MTCC culture center. Google to know more. Or you can avail them from any universities or research institutions who are working with it in your state

Improper sterilization, add antifungal compounds.

@@Dr.Rakesh.B actually I initiated 6 flasks got yeast contamination in one flask..others are good ... I do sterilization process regularly.. in recent days I got contam.. can I ask u doubt by calling sir.. contact no if possible
