Rye Berries are my favorite grain to use to generate high quality grain spawn.
Soaking the rye grain overnight is recommended to germinate any endospores that can withstand sterilization temps. Once germinated, they will be susceptible to the sterilization cycle.

I find it best to boil the water / turn off heat source and then dump the soaked/drained grains into the very hot water stirring periodically for 20-30 minutes. Drain and lay out of a towel for and let the steam evaporate the moisture.
Sterilization: 15 psi 2-2.5 hours
Shaking the jars after they have cooled down will help to distribute moisture.

Guys... Spreading the grain out into a mushroom is a necessary step. This promotes a emotional connection with the grain and provides happy grains, which in turn promotes happy mycelium. DO NOT FORGET THIS STEP!

Hey thanks for the video do you have any information on spawn grain to substrate ratios? I don't know how much grain to prepare? I have a huge mono tub its 35 inch long, 11 inch wide and will be 2.5 inch depth of my substrate. Im new and lost with this particular aspect. Thanks

Your videos a great! Thanks so much for producing them. I purchased some of your cultures and I'm beginning to sterilize substrates for inoculation. One question: you don't add any water to the jars after drying the rye and before sterilization? Does the grain get back moisture in the autoclave? In the Stamets book they add 2/3 - 3/4 c. Water. But they also don't cook it before, so this may change things. Thanks for your work!

I don’t add any water after drying. All of the moisture the grain needs is absorbed during soaking and stirring in the near-boiling water. There are many different techniques but this one works great for me still.

Once your done with this can you take the jars out of the still air box and leave them anywhere? I was wondering if when using micropore tape you need to cover the lid with aluminium foil or if the micropore tape is enough. Thanks for the vid!

I think it's good to add coffee grounds to it too, save some old coffee grounds for a couple days and dumb those into your bucket along with your grains and gypsum, it will make a world of difference.

@@judyhowell7075 Just a pinch between your thumb and finger is all I used, I used the pellet type because I couldn't find powder anywhere and it worked great. I've heard it's not really precise so just a pinch or maybe a half a teaspoon something like that

Before putting the jars into the pressure cooker, do you have the lids on tightened all the way or just "finger tight" to let it breathe? I've heard both so I'm not sure, great video btw!!

I leave them a tad loose, it not a big deal if you forget. You can add an extra piece of micropore tape if it becomes blown out after pressure cooker.

Call grain and feed stores, or grain elevators, make sure to ask for “rye berries” not “rye”. Or you can try using wild bird seed or oats.

rice is a junk grain spawn it is better ground into a powder and mixed with vermiculite. I have attempted using rice as grain spawn a million times, it is not good. I have made it work but it is more or less junk

Any one know where I scan get liquid culture psilocybe cubensis at bought spore syringes about four weeks ago and did everything and still no sign of colonization I did research and heard liquid culture or agar if faster and easier and I’d like to give it a go

You can try that but it is not recommended. Grain is highly nutritious and needs to be sterilized under pressure.

If you use popcorn kernels instead of rye you can use the preserving machine. Rye needs pressure

Hey brotha, quick question. So Inoculated brown rice grain spawn about 3 days ago with Lions Mane. 3 of the 5 jars are showing mycelium growth pretty strong spots and 2 of them I can’t see them doing anything. Is this normal? Temp between 63-70 degrees F, humidity between 80-90 RH

3 days is a very short period of time for lions mane. The fact that you are already seeing spots on a few of the jars is good. Give them more time (1-2 weeks). Also lions mane mycelium is very thin and hard to see at first.

How do you keep the grains dry for cultures that colonize slowly? Wet rot eventually gets to mine before all the grains are colonized.

After straining the grains from the hot water the steam helps evaporate moisture while laying out to dry. I mix up the grains 3 times every 10 mins while they are drying on the table. Also it helps to use 4-5 ml of liquid culture or a whole agar plate full of mycelium cut into many small wedges to inoculate the grains.

Depends what kind of air filter you have in the jar lid. You don’t want water wetting micropore tape. Excess water entering the jar can also cause the moisture content to exceed normal levels.

once the mycelium grows in mason jar how would you get it out? I assume it would be solid like a brick. Would you break it up with your hands?

Yeah you can smack the jar on a bike tire or something to break up the grains. I use bags and those are easy to break up.

About 5/10% of my rye grains split open during the process. I know now my temperature was too high. I’m picking out as much of the split ones as I can. Should I start over or do you think I’d be okay?

I usually pick out a hand full of bursted grains and it works fine so I think you will be ok. You should boil the water first and then shut off the stove completely and then dump the grains in while stirring frequently for 20-30 minutes. That should help cut down on bursted grains in the future. Sometimes while laying out the grains to dry, the steam will dry the grains too well so I recently started rinsing the cooked grains with cold water to eliminate the excess loss of moisture from steaming, this helps retain moisture if the grains become too dry over time.

Yes store in a cold room, but the grains tend to dry out over time pretty fast so I would use before 7 days

On your cooker, do you have psi meter or how do you know if the pressure is 15 psi. And can you inaculate the jars with spawn plugs/dowels?

My new pressure cooker has a a psi gage, but some do not have one. There are two weights on the pressure cooker in this video that creates 15 lbs of pressure. No matter what kind of pressure cooker you use, you need to use 15 psi for 2.5 hours.

@@ziggysplace3858 Yes that is normal, you should shake the jars to mix up the moisture right after pressure cooking. If there is still pooling water after shaking then that’s bad. Its good to use tinfoil to cover the jar lids but sometimes the filter still gets wet. I usually put the jar in front of the flow hood and let the filter dry if it got wet or just rip it off and put a couple new pieces of tape.

You are fully cooked, the grain will be too dry. You could try to soak the for a couple minutes and dry again but I would probably start over

Of all the grain spawn preparation methods, I like yours the best. I have tweaked it a little by i.e. adding some gypsum. I then use large grain bags which after sterilization I inoculate with some Uncle Ben’s tek material. This multiplies the colonization (by 3-4x) which I then multiply further (by 2-3x) by adding the grain bags to coco coir + vermiculite monotubs. I really like and use the relevant bits of the revised Gordo-tek by Rick James ru-vid.com/video/%D0%B2%D0%B8%D0%B4%D0%B5%D0%BE-Pm1FgFFzQd4.htmlfeature=shared and Boomer Shroomer’s technique ru-vid.com/video/%D0%B2%D0%B8%D0%B4%D0%B5%D0%BE-Pm1FgFFzQd4.htmlfeature=shared. I tend to be liberal with the use of hydrogen peroxide during both the preparation and the growing processes and seldom get contamination. My own extra tweak is to use a small tanning lamp (Fiji Sun) which I turn on for 10 minutes 2x per day - I haven’t measured it but the growth seems faster and the mushrooms more potent with a little sunshine, even if just imitated ☀️🤓

@@HasF.4kZ It’s as simple as asking a question. Liquid culture is stored on a shelf in a room that is 70-80 F while colonizing, once enough mycelium has grown the jars are moved to a. Cooler room 60-65 F to stay for up to 6 months. Grain spawn is basically the same, but should be used within 3 months.
