Calibrating equipment is when you adjust or mark the equipment so that it can be used to measure in an accurate way. In this instance, with microscopes, when we calibrate it means we are working out exact what each division distance represents at each magnification in the eye piece graticule (the ruler inside the eyepiece). This enables us to then accurate measure the structures we are viewing under the microscope.

miss so does 1 division on a stage micrometre always = 10MicroMetre

Hi, one thing I don’t understand is how does a division on the eyepiece graticule equal 5micrometers and not 20micrometers? I’ve been thinking that as 2 divisions of the eyepiece go into 1 division of the stage micrometer, then we need to times by 10 to get the eyepiece division micrometer? 😅😅 I can’t seem to see where I’ve gone wrong

In exams, when the diagram sometimes has labels (etc. A, B, C, D) on the organelles, but the organelle is not clear, or could be one of 2, how would you best figure out which one it is?

I think this is asking you to calibrate as shown in the video, I imagine the question has a micrograph image with it and a scale to calculate this?

Hello, at 15:56 you are explaining that the nucleus is 13 divisions long, but isn't it 13 sub divisions long. In the previous slide we deduced each division of the eyepiece graticule is 5 micrometers, so I was just wondering if the answer for the question on 15:56 isn't 65 micrometers, but a value a lot smaller due to the nucleus being ~1.3 divisions across (instead of the sub divisions). A great video though

Hi MadHouse, Miss Estruch is correct becuase when she mentions one division, she means 1/200 If the stage micrometer is 2mm long, therefore 2000 micrometers, each division is 10 micrometers. If two divisions on eyepiece graticule fit into one division on micrometer, one division on graticule is 5 micrometers. The nucleus is 13 divisions long there around 65 micrometers. Miss Estruch is correct. Thanks :")

If the stage micrometer has 100 divisions and each division is 10um then the stage micrometer is 1mm long not 2?

Hello, Yes, well spotted, my image isn't to scale. Stage micrometers are usually 2mm long, but I haven't got an image quite to scale, and it is only showing the first 1mm. It would extend another 100 divisions to make the full 2mm.

How do you work out the volume in diagrams on light microscopes?

this would be testing your geometry skills. For example, if viewing a spherical cell and you need to work out the volume then you need to measure the radius and then use the formula for the volume of a sphere. This might help ru-vid.com/video/%D0%B2%D0%B8%D0%B4%D0%B5%D0%BE-qtEpAJ7NK_w.html
