How to calibrate the eyepiece graticule using a stage micrometer so that it can be used to take measurements of specimens under the microscope Check out this video (theory / worked example): • Measurement Theory
Before the video, I wasn't sure if you can zoom in or out after you calibrated since I thought it would change your conversation number. This helped clear that up :). I have a practical assessment on this tomorrow for my A-level Biology course so this was a life saver :).
Okay so you choose the magnification and focus it. Place the stage micrometer under the lens. Line up the the piece graticule where two lines meet. Divide by how many eyepieces there are to work out how much 1EPU unit Is worth - corresponding to the notion that each small line is equal to 0.01 mm and altogether 1mm. Measure the new sample without the stage micrometer and use the eyepiece graticule to measure how many units it is and times by the use of ratios from the size of one eye piece unit?
Dear MrMyfra. If you feel that there is something wrong (or inaccurate) wtih this video please let me know. Always interested in feedback on how things can be improved.
I want to point out a mistake. You said that we multiply 0.099 by 3 to get around 0.3 mm, whereas we must multiply by 30. And the same goes for the other cell.
