Okay got it, so C3 is cleaved into C3b and C3a by C3-activator C4b2a which makes C3PO into C3bBb using a Ceetec p40, which means black moves his bishop to C3 and this is how it all works

Took me a while to understand what you meant by checks. If you would understand the teaching method, it makes much more sense to see how the alternative pathway is related to the rest in this way. Its a wiser idea also not to give your address to the public. Thanks Armando

Hi Armando, great video but you have a mistake in C3-convertase. According to my literature and my professors c3 convertase is actually c4b2b. I would really appreciate if you answered me with your opinion and resolve my doubts. Thank you for making my life easier!

+Rif KLAIMI I believe that is because C2a was traditionally attributed to the large product of C2 cleavage whereas every other C#a is the small product. One of my books (Janeway's Immunology) tried to change that around but it didn't really take off.

You said that the alternative pathway is the last to begin since the C3 convertase made by the other two is needed. Now am I taught that first it's the alternative, then the lectine and final the classical, because in classical antibodies are needed and these have to be synthesized first. Also since C3 can react with water to make iC3, next CB attaches and gets cleaved. This leaves iC3Bb (also a C3 covertase). Showing the alternative can do it all on its own. Any thoughts?

I think this material is outdated. The Alternative Pathway is actually the first to be activated because C3 has a highly reactive thioester bond that spontaneously reacts with water to make iC3 when it is near a pathogen. The iC3 then binds factor B and then factor D comes in to cleave B into Bb and Ba to give rise to soluble C3 convertase -> iC3Bb. The soluble C3 convertase iC3Bb is now able to cleave and activate C3 into C3b and opsonize the pathogen. The Lectin and Classical Pathways follow in sequential order after the Alternative Pathway occurs. The Lectin Pathway requires an induce response from the body to make more MASP-1 and MASP-2 to activate C4 into C4a and C4b. Lastly, the Classical Pathway occurs last because it requires an induced response from the body to produce Ag-specific IgM/IgG or C-Reactive Proteins for the C1q heads to bind.

Btw, C1qr2s2 complex doesn't just bind to C4b2a. It gets activated when it binds to an antigen-antibody complex causing it to activate C1s and cleave it away. The activated C1s will cause C4 to cleave to C4a and C4b. The C4b now has a small part missing, to which attached C2 and cleaves itself to C2a and C2b...and removes the C2b part. There we form the C4b2a complex

Isn't the alternative always active at a low level? If I understand correctly, C3 can autolyze forming C3a and C3b, which binds non-specifically to cell membranes. The sialic acid in host cytosolic membranes inactivates (somehow) C3b. Bacterial cytosolic membranes, lacking sialic, become decorated with C3b allowing the cascade to progress.

For the alternative pathway, there is only one C3 convertase created: Facor D is aprotease that cleaves factor B in to Bb, which forms the final C3bBb complex. The Properin is an enhancer/ stimulating factor of the c3 convertase to create more fixed C3bs and therefore enhanced opsonization... p.s. where can I find the videos that are specific to each pathway?! :)

But doesn't the Alternative pathway activate first? This video makes it seem like the Alternative pathway is last but I'm pretty sure it's the first one to activate. There is C3 regularly in the blood and it doesn't need the other complement proteins to make its C3 convertase, just Factor B and Factor D.

Mr.Armando,l beleave that theory are good,but when you're an elderly person you need something more practical.If your bone marrow has been destroyed by a chemotherapy and you are in a big problems,you needed someone to teach you how to survive daily!

Just one question. Whats the difference between viral infection and bacterial infection. My friend told me it is T cells take responsible to against bacterial infection, while B cells are for viral infection. Why?

In the Innate Immunity response besides the barriers is inflammation,witch is a key step for the adaptive immunity ,without inflammation adaptive immunity would not function.
