When using restriction endonucleases, the production of which of these ends are critical to recombinant DNA technology? complementary double-stranded ends, double-stranded sticky ends or complementary single-stranded ends? Thank you so much

do you use the same restriction enzyme to that cut the plasmid to cut the DNA that contains the gene of interest?

But when this sequence ( restriction enzyme sequence) will present in other bacteria whats happen then, because its have ability to cut the specific sequence ,as you said

EcoR1, and other restriction modification systems, are deleted in laboratory E. Coli strains.
