Acne is a multifactorial inflammatory disease that affects the skin's oil glands and hair follicles. Current acne treatments target the three main factors leading to breakouts: excess sebum production, bacteria growth, and clogged pores. This pharmacology lecture covers topics such as pathophysiology of acne formation, overproduction of sebum, follicular hyperkeratinization, the role of P. acnes bacteria, genetic & hormonal influences (the role of estrogen and testosterone), mechanism of action and side effects of common acne treatments; retinoids, pertinent antibiotics, and hormonal therapies. Drugs mentioned include; Tretinoin, Adapalene, Tazarotene, Isotretinoin, Sulfamethoxazole, Trimethoprim, Clindamycin, Doxycycline, Minocycline, Erythromycin, Azithromycin, Benzoyl Peroxide, Spironolactone, and Oral Contraceptives; Ethinyl Estradiol, Norgestimate, Desogestrel.

When I started using retinol I had no idea it could help with acne; I was using it to even out my skin tone. When the breakouts stopped I was pleasantly surprised and incredibly thankful.

This acne guide *AcneCureOnline. Com* was very informative and helpful. I often go through gentle adult acne and this guide discusses all the possible solutions for mild to severe acne whether natural procedures or otherwise. I prefer the natural way of treating them and I found out on this guide the treatments that work efficiently.

My dermatologist is incredibly reliable and a respected professional, but she just took 1 look at me and prescribed 1) Epiduo (Adapalene + Benzoyl Peroxide ) 2) Clindamycin 3) Oral Hormonal Therapy 4) Blood Tests to change Hormonal Therapy to a stronger one (the Pill) if needed WITHOUT ANY EXPLANATION, I wish she told me what these products are supposed to be doing. Thank you for your explanation, now I understand I need to be patient with redness, irritation, itching, peeling etc, as they’re a normal part of healing!! :D (I’ve only been doin this treatment for a month, but I can really see my acne calming down and reducing to small spots where it was less severe, now I just need to le patient and wait for my skin to regenerate like it’s supposed to)

This was so informative and well presented! As someone who struggles a lot with acne I really appreciate being able to learn about it in a more scientific way. Would you ever consider making more skin care videos? For example, how retinoids work for anti-aging or maybe on different sunscreen filters and their mechanisms :)

I never had to deal with acne during puberty, but I’m now in my mid/late 20s and my face just doesn’t want to cooperate! Now I understand a little more about how my tretinoin and clindamycin work together to treat my acne. I just have to be patient...

ive been using both for years and to be honest it works and the zits come back again cause they gain resistance. its all in the genetics in my opinion but it definitly helped prevent worst case acne

This is exactly what I needed. My dermatologist just prescribed me to use facewash with benzoyl peroxide, clindamycin and tretinoin. Thanks for explaining it in a way that makes sense!!!

Wash my face twice a day so ill explain morning and then night CeraVe 4% benzoyl peroxide and clindamycin wipes in the morning with CeraVe 30 spf moisturizer. At night, wash with CeraVe 4% benzoyl peroxide and then after i use the tretinoin mixed together with CeraVe night time moisturizer. I had to work up from once every 4 nights tretinoin to every night slowly it took a month to get to where I was using tretinoin every night

So you cant use both prescriptions at the same time so it’s clindamycin in AM and tretinoin with moisturizer at night

I am in my 40`s and just began with adult acne. It got seriously bad!! I didn`t experience acne before. After applying this acne guide *AcneCureOnline. Com* for 2 weeks, I observe obvious effects. I am trying this treatment for 2 months now and have become comfortable once again. .

There Is an Overload of Conflicting Information and Often Ineffective Advice on How To Treat Acne. I had terrible acne from my teens until my 30’s. Changing my diet is the only thing that worked-I went paleo and the acne is gone!

meena mee Hi Meena, we are all different but for me my diet was the culprit. I did an elimination diet called the Whole 30 for a month then I transitioned to Paleo. Basically I cut out dairy, grains and sugar. I think dairy mostly added to my acne trouble. Acne was so frustrating for me, it got super bad after I had my first baby but thankfully the diet cleared me up after years of trying all the topical creams. I wish you so much luck on your journey.

Hmm this is somewhat confusing on the retinoid section. It's mentioned and it's true that retinoids have to be converted to retinoic acid but the examples given I believe aren't part of that situation. Tretinoin is already bioactive retinoic acid. Adapalene and Tazarotene are both third generation retinoids that are already bioactive even though they're not retinoic acid, but they do bind to the same sites. I was hoping to get some information on what the key differences are in the pharmacology between tretinoin and adapalene.

This also happens to women that are menopausal see them goes up oestrogen is gone I ended up with folliculitis and prurigo nodularis. My doctor said this is my new reality this horrible crap leaking on my head when in fact I asked him to put me on Accutane because I had used it years before and it seems to be helping

Khalled Alves Isotretinion is taken orally and used to severe acne. Tretinoin is taken topically and use for mild to moderate acne.

This is so good and informative. Great video, clear and aesthetically pleasing lol.. I wanted to know what I was putting on my face. I stopped using adapalene and my acne came back. What can I do now till I take my next dermatologist appointment ? Is there a way to stop them for a few days? Im not able to plan my dermat appointment because of busy schedule and cant risk having acne back coz I'll be having events next week.

In the cells, retinoic acid enters the nucleus where it binds to heterodimers formed by two different classes of nuclear receptors: Retinoic acid receptors (RARs) and retinoid X receptor (RXRs).

Excuse me Sir, I want to ask for my research paper. So when we give antibiotics, what happens to the bacteria? Are they totally killed or reduced or deactivated or what? Because as I know, P. acnes are naturally exist in every human skins. Thank you

P. acnes is part of our skin's natural microbiome. Some but not all strains of the bacteria promote acne-causing inflammation. P. acnes colonizes primarily sebaceous glands and hair follicles but it may also be found in the mouth, nares, genitourinary tract, and large intestine. It is able to recolonize quickly after a round of antibiotics.

@@ibibercerita antibiotics commonly used in treatment of acne such as tetracycylines are inhibitors of growth (bacteriostatic) rather than killers of the bacteria. see my video about the pharmacology of antibiotics for more detailed explanation
