Neither Lois or Clark acknowledged feelings for each other yet, but Maxima knew there was something because her powers were unable to overrode the connecting between Lois and Clark. Which is why Maxima tried to kill Lois
They've had a similar conversation before in season 5, except Lois was the one wondering about meeting the one and Clark is reassuring her that she would. They are almost in the same positions in the barn for both scenes Search for "Clark Kent & Lois Lane: "I promise someday you'll meet someone even more special..." and compare scenes
look at Lois's reaction and hesitation struggling with an answer before answering Clark when he asks "what if my soul mate comes along and I'm to blind to see it"
These two were still on separate journeys at this point. You can definitely see something there between them both. The Lana Saga wasn’t over yet. They had to finally write her out to get to Lois.

maybe because Clark and Lois just fell in love when they start working and spent almost everytime they had together. the tv show showed her before the right time...So I guess they just tried to make things right when they decided to stick with the idea that they should fell for each other in daily planet.

He’s worried about not finding his soulmate about not being able to find her even if she’s right in front of him; hence the irony. She’s his soulmate, her being able to tell in a split second he was really Clark after he and Clark Luther switched back proves that.
