Welcome to the fifteenth episode of season 2 of The Sci Guys. In this episode we are extracting the DNA from a strawberry. We will show you how to mix a solution that will take apart the strawberry cells and release the DNA from inside them. Strawberries have a large amount of DNA compared to humans and we can easily extract it and pull it out of our solution with tweezers. Strawberry DNA looks like a cloudy ooze.

I typed out their method for people who need it for school: 1. Mix the water, salt and dish soap together in a bowl. 2. Place the rubbing alcohol into the freezer to chill. 3. Put 3 strawberries with the stems removed into a Ziplock bag, measure out 3 tablespoons of the previously made solution and pour that into the bag also. 4. Seal the Ziplock bag closed, remembering to remove the air and crush the strawberries until they are a fine, foamy paste. 5. Strain the strawberry pulp into the beaker, using a spoon to push it through. 6.Remove the rubbing alcohol from the freezer and measure out half a cup of it. 7. Gently pour your alcohol into the strawberry mixture, making sure that they don’t mix and that it just makes a fine layer on top of the pulp. This will help the DNA separate from the rest of the strawberry mixture. 8. Use your tweezers to remove the strands of DNA from your beaker.

How much saliva did you use? Probably up to a cup to make your DNA really visible, haha. I only get a dot of DNA from about 2mL of saliva

I did this experiment at school last year under standard curriculum, and now we're redoing it under honors; I've yet to see the changes implemented for this year in contrast last year, but last year's was pretty similar to this experiment. I don't think our teacher trusted us the tweezers ...

Hey Justin, I couldn't reply to your comment directly, probably because your comment is linked. You can do this method with a lot of different fruits. The thing to remember is that if the fruit isn't octoploid you will require much higher quantities of it to get a significant amount of DNA. You might be able to spread the DNA out on a slide and look at it under a strong microscope. You may not see much, but you might see something.

I had a problem with making the layer of alcohol. It made it, but it formed beneath the strawberry mixture. Do you guys know why? (I did pour it gently)

In my practice, I put the salt first to smash the strawberrys and then the detergent, actually, i used pineapple juice and i don't know why and i don't find any information of that :'v The result was a little bit of DNA. Could somebody explain that? (notice my terrible english)

Silly question- but would this method work with other materials? Food wise, but also granulated substances such as sand, dust, crushed rock etc. I have to perform this experiment to prove that the provided samples that are not necessarily 'alive, may contain living organisms...not sure if this will work.

The alcohol needs to be cold because it helps increase the yield of DNA. Cells contain enzymes in the cytoplasm that help destroy viruses but protect the DNA. Hot water has the potential to damage DNA by accelerating enzymes that break it apart. DNA is typically protected from such enzymes by the nuclear membrane, but the soap added previously destroys it. Cold alcohol helps the DNA precipitate faster.

You can use a high proof alcohol (ethanol) and fresh pineapple juice to do this as well. Skip the salt and soap, and instead freeze the strawberries and then let them thaw. Ice crystals will break up the cell membranes (much like the soap). Put about a cup of fresh pineapple juice (not pasteurized) in a baggie with about three strawberries, and squish. Strain the strawberries, and then pour the adult beverage the same way that the rubbing alcohol was done here. The DNA will separate, AND the entire concoction is now made of 100% human consumable ingredients. The Pineapple juice replaces the salt, and the freezing of the strawberries eliminates the need for soap.

Chelssums You can keep it, or observe it under a microscope. Put it in a test tube filled with the remaining alcohol and keep it, or you can let it dry out.

Соль помогает выпасть в осадок все ненужное - оболочки клеток, ДНК будет в верхних слоях раствора. В воду добавляют соль и жидкое мыло, этот раствор выливают в полиэтиленовый пакет с клубникой и давят ее. Через 10 мин. процеживают раствор. В сок клубники добавляют охлаждённый спирт. При добавлении спирта происходит отделение ДНК и затвердевает как полимер (как сопли). Затем вам нужен будет сканирующий электронный микроскоп (Scanning electron microscope), чтобы рассматривать ДНК.
