Welcome to the third episode of season 2 of The Sci Guys. In this episode we will be investigating how to make a homemade petri dish and how to mix together a culture medium that will encourage bacteria growth. We will show you how you can explore bacteria growth and petri dishes at home with a bunch of simple household items and at the end you will have a homemade petri dish that any bacterium will want to call home.

Mixing Bowl

Plastic Wrap or Lid for the DIshes

Lab Coat or Apron

James Collins Sweet!! I hope it turns out well. If your growth medium is turning to liquid at room temperature, then retry your mixture with an extra package of gelatine or reducing the amount of water in your mixture to make a more resilient medium.

My suggestion for a good Petri dish prep video would be to have a couple of kids doing all the work with the science guys looking on.

Hello (: I'm a long time fan of you guy and I have a question, what did you do to substitute a warm humide environment for the Petri dishes to grow properly? Any suggestions on what I should do to get the best results? I LOVE SCIENCE so don't be afraid to use scientific terms hahaha, thank you!

Kemija je zakon You are correct, there could be some fungi growing in them as well. Especially when we consider what could be growing in the dog's water dish. The dish that had the mouth swap is probably the least likely to have fungi growing with the bacteria but there is always the chance of contamination.

Ariel Barton The stuff growing in the samples we grew can be disposed by cleaning it with hot water and dish soap. It's not much different than cleaning your dog's bowl or a dirty dish.

If I put something like a mini glass substrate/squared cut paper onto the bacteria medium and waited for several days will the medium cover the substrate?

Thank you for this information. I wanted to test my water supply. Our water supply comes from a special water tank that is contained under the house (cold, dark, and deep tank 15000 gallons). Our roof top collects rain water (it rains allot here) and is deposited using a drain gutter and pipes into the

Hello. I live in Arizona. I want to try this with my 12 yo daughter. But humidity doesn’t exist here unless it rains lol. Should I allow the bacteria on the Petri dish to grow in the restroom? It’s dry, dark and some humidity when we shower?

+MegaMackproductions If you want to culture yeast then I would probably recommend using Agar instead of gelatin. It will give you a material that won't liquefy as easily

I home school my 5th grade daughter with the K12 program. There are many experiments listed in their Science program guide, and often times, it would be overwhelming for us to try to do them all. Your channel has been a great resource for us to sometimes watch what happens in a more timely and safer fashion. I did recommend your site to our Idaho K12 school. Maybe you could get a hold of K12 nationally and work out a deal make experiment videos for them (they work with many off site places). I think you guys have something special that could really be beneficial for students out there and profitable for you. Good luck!

It does not matter how long you've boiled the water since you've poured it in a non sterile bowl, mixed the non sterile ingredients with a non sterile wooden spatula, again in a non sterile measuring cup and lastly in a non sterile 'Petri' dish. And you've used a non sterile q tip. To make sure that grown colony is your dog's 'bugs' or ones from your mouth, everything has to be sterile at all times, minimal exposure to air. ;)

@@stupidstickmen84 expose to the air is still gonna cause contamination. the only way to really do this with sterile technique is using a pressure cooker and a flow hood or no pour tek, we use this in mycology because we need super sterile conditions to germinate spores and grow mycelium

Julian Mory Hello Julian, to be honest we just placed our petri dish in our basement in a room that was away from sunlight. Bacteria and Fungi will grow faster in humid and hot environments but they will still grow, just slower, in the normal conditions in your home. You could use a heating lamp to increase the temperature around the petri dish, but I wouldn't point it directly at the dish, just in a direction that will warm the air around it. For humidity you could use a humidifier that some people use when they are sick to increase the moisture in the air. Also you should look at the settings on your account to allow people to reply to your comments because right now I cannot reply to your comment directly.

How can we get a control bacteria to test the effectiveness of cleaning agents for a science fair project? If you cant or dont want to buy actual petri dishes, wide mouth half pint Mason jars work fantastic, both as an improvised petri dish, and also makes a very good Crystalizing dish

We should come up with a way to convey quantity of something by weight and/or volume in a form that dosen't differ depending on place, person or time . Two packets gelatin (not that crazy Knox gelatin packet size is standard now but there are two sizes that look identical except for the size) and one packet bouillon ( never seen before, so beef base in jar is out). Sigh...........

Can you grow bacteria without any of this stuff? E.g. a strong growing bacteria or fungus... would it simply grow in a bit of water and sugar in a glass dish?

This is called a liquid culture. Although typically you need extra nutrients, not just food. (Just like humans can't survive just off calories, each bacteria will typically require some micro/macro nutrients.) There are plenty of variations on different growing mediums for liquid cultures. But you loose the ability to identify the bacteria via the morphology.

violet Boil the water first then add the other ingredients. You can get beef bouillon at any grocery store. Also you don't have to use beef, you could also use chicken bouillon. It would just look a slightly different colour.

hello comments, I have an EXTREMELY urgent question and I need an answer ASAP. I am doing this project for science fair, and I am on day 4. I see ZERO signs of bacteria. I followed the directions perfectly to the tea for everything, yet nothings growing, I just checked on it. So my question is: When should I start seeing results? Like around what day? My science fair project is due soon and I am so afraid I have to do a whole new project and get a bad grade. Im freaking out I wanna cry right now. Anyways, if you do answer this, thank you so much!

MICROBES AT HOME? A REAL CHANCE FOR ABLE PEOPLE TO GET WORLD RECOGNITION. Before anything else care should be taken to stop FECAL DUST forming. Hospital Acquired Infection: Hospitals try hard to kill the microbes in the wards, but give little attention to eliminate the microbe SOURCE, which is the feces residue, left after incomplete wiping (it always is somehow), that through drying and friction it turns out to the lethal FECAL DUST. Have in mind that hospital patients are usually, debilitated old people, or having mobility restriction, unable to wipe properly, particularly, if there are piles or hair. HOME AIR AND FOOD CONTAMINATION: The same process is taken place here. UTI: The difference in UTI is that fecal dust and the contamination microbes come from one’s own fecal residue. Many suggest front to back wiping, because they think that microbe transfer is done only by direct contact. It is a big mistake, as they ignore the transfer through FECAL DUST. “Why no public research health institution does a simple research about the LETHAL FECAL DUST in hospitals? It could save millions of deaths and trillions of $, on hospital function cost, or it is exactly that cost reduction is the reason that it stops them to do it?” ANY BODY CAN DO HIS OWN RESEARCH WORK TO PROVE OR DISPROVE WHAT I AM SAYING AND GET WORLD RECOGNITION!!!

Use agar-agar. It's inexpensive and the microorganisms can't metabolize it.

Boil water for two minutes to make it sterile. 😂 (Except for spores and TB or any other mycobacterium) Then insert dirty wooden spoon to stir. Lmao. It’s a cute experiment but I’m dying at the methods used. 🤦‍♀️🤷‍♀️💀

i would say so, it has sugar, only thing i suppose is that the colouring may make the bacterial growth look slightly different. try it!!

@@zirconium2014 use unflavored or remove sugar from recipie. Should work but may give some weird colored bacteria cuz of eyes ;)
