@@RandomGuy-lu1en In that case, best to avoid increasing it. Some age-related changes are pathogenic, others are adaptive. In the case of NAD+, the higher the level the greater the chance of terminal cancer. This is because NAD+ is required for energy and cancer cells need energy more than healthy cells (it does not cause cancer but it will feed it). That being the case, it is far from clear that age-related NAD+ decline is pathogenic.

@@antonystringfellow5152 I don't agree here. With the high NAD promoting cancer in older age. Not necessarily that is. You need to control what the NAD+ does in the body - you don't wanna feed it to CD38 or some dubious PARP enzymes - and you want a healthy gene expression so that NAD+ is used the right way - for that you need many more nutrients like useful phytonutrients that control the gene expression, gut bacteria, the enzyme activity (including NAMPT). So, yes, you can get more inflammation and diseases if controlling for NAD+ only. But you certainly wanna push NAD+ but make sure you eat healthy otherwise, or correct other bottlenecks while raising NAD+.

@@antonystringfellow5152Any theoretic benefit for cancer cells is more than offset by benefits to immune system that are a check on cancer. There is no evidence of cancer increasing in people who use NAD booster and the opposite appears to be the case.

Norway study (2018, Midttun, et al) showed plasma trig of about 3.6 micromol/L for 2 to 4 cups of coffee per day. zero cups per day was 0.3 micromol/L in plasma. 1 cup per day 1.6 micromol/L.

It certainly is useful when you also correct the immune system for them - that is mostly achieved via treating the microbiome - so better use legumes and other seeds for trigonelline, at least partly, and use more whole plant food in order to feed the immune cell (which require plant fiber metabolites). Legumes are a nice 2 in 1 anyways - you also get loads of natural folates from there. Folates that closely participate in NAD+ cycle and trigonelline.

thank you very much, that was very exciting!! On question left for me: the stability of trigo in plasma is quite high, and at the same time the NAD levels rise. Could we conclude that it is not directly converted into NAD via Preiss Handler, but rather has an upregulating effect on NAPRT? The stability is probably very important to reach all tissues, not just the blood compartment. Regarding plant sources I found an interesting paper: Hammel e al, 2013 ."N,N-Dimethylpiperidinium (Mepiquat) Part 1. Formation in model systems and relevance to roasted food products" ...which is not directly about Trigo . Yet they measured the Trig content of various seeds, and found that Trifolium incarnatum, Crimson Clover seeds contain even more Trigo than green coffee beans. Moreover, the whole plant is edible, and it is known that plants synthesize Trig for seeds and young tissue. ..and it is very cheap: 25kg cost 3$

Hi Michael, Have you found any data on methyl-niacin vs niacin? It's not really available to buy, and the little that is - very expensive! (Less that 3X of the effect vs niacin doesn't justify at the moment from a practical perspective IMO. )

Hi Rimon, not yet, NAD data should arrive this week. From daily chickpeas, I was able to increase methyl-NA by about 20mg/d, but whether that's enough to make a dent on NAD, we'll soon see. Note that 60 mg of nicotinic acid raised my NAD from 20-25 uM to 39 uM, so I may see a bump with the daily methyl-NA increase. If it doesn't, I just bought fenugreek seeds, which have a lot more methyl-NA than chickpeas, and will be the next NAD experiment!

on amazon there is green unrocasted decaffeinated coffee beans available how much trigo do they contain tho? without messing with one's caffeine intake, considering trigo is solvable in alcohol, one could toss those beans into a wine bottle and just wait until one had trigo-wine this trigo wine then could be used as a (pre-meal) apéritif/digestive bitter to increase bile acid flow, its like a win-win-win combination of things

Thanks Michael. Appreciate these presentations/discussions with scientists reviewing recent work. I have 1 cup of coffee plus 1 cup of decaf coffee in the morning before exercise and walk. Wonder whether there is benefit to exercising with a transient increase in NAD+ , beyond that provided by exercise, assuming my coffee increases NAD+ (although data shown here seems to show coffee, via caffeine marker, does not increase trig).

Thanks @peterz53. Although there is published data that exercise increases NAD, I'm not sure that's always the case. Anecdotally, my 20-25 uM level is age-expected, and I'm on 2 90-minute, whole-body workouts/week. Decent fitness, but low NAD...

​@@conqueragingordietrying1797thx Prof LUSTGARTEN for this exciting discussion.As for exercise and NAD it has been shown that 14000 steps per day increase NAD of old people to the level of individuals in their 20 .Furthermore , from the present video we can conclude that , for trigonelline to be effective in improving force and muscle it should maybe combined with exercise.Maybe drinking coffee just before exercise, especially high intensity exercise or sprinting,could be beneficial.Anyway, it's great to know that centenarians have high level of trigonelline and that molecule promotes muscle résilience.

Exercise does not increase NAD+ directly. In most part (there is others) it is supposed to increase the activity of NAMPT enzyme which is the enzyme that turns the niacinamide back into NAD+ (so the salvage pathway and where most of our NAD+ comes from constantly - so THE MOST IMPORTANT) - it does so partly via AMPK pathway - AMPK is the energy sensor, so to speak - you energy levels drop by exercise or fasting and not eating, then AMPK is raised and it, among other things, activates NAMPT - so, in my theory you get the best NAD+ levels (might not immediately but it activates the NAMPT for the later of the day) while exercising with empty stomach (it actually simulates searching for food, hunting for animals, in ancient times), and while undergoing other NAMPT-promoting solutions (plant phytonutrients etc.).

If methyl-niacin is 3x more effective, bro science tells me to triple my flushing niacin NA dosage to match methyl-niacin. Sticking with that until further notice. And take TMG to protect my methyl groups

OT: Suggestion for a topic to explore - the relationship between seasons, RHR & HRV, and mortality. Does RHR increase / HRV decrease in winter just because of temperature? Or is there something else involved, like sun exposure or geomagnetic flux? Are people better off in terms of health and longevity by moving to a warm, sunny climate (typically lower latitude) in retirement - or do they just need to stay active? I'm sure this topic would be of interest to many people. Personally, I've noticed a reduction in chronic pain when I can get out in the sun - I can feel it almost right away. In the summer I was able to do 20 reps of 25 lb Romanian deadlifts - now I can only do 6 reps and stop because of hip pain.

Hey @rhyothemisprinceps1617, the only season that affects mine is early summer (late May-June), as seasonal allergies mess up HRV and RHR acutely, but if I'm not careful with allergen exposure, it can be for a few weeks, which isn't fun.

But I'm sure you'll be doing a test to figure how much it increases nad 😁 What happens if you take fenugreek with supplemental NAM or nicotinic acid in nutritional yeast?

I drink coffee, and its trigonelline properties seem promising, however i feel like the undiscussed issue with coffee is the fact is excretes sodium, which in turn causes u to need more sodium --> and then consume more sodium. i LOVE coffee but i am slightly skeptic😖

What about green coffee bean extract as a source of trigonelline? Would that be an option? Any way to guesstimate the trigonelline content? What was a reasonable(guesstimated) dosage for humans? I had a hard time understanding the audio.

There wasn't clear dosage recommendation mentioned. Michael seems to predict it may be a low amount, a similar to NA that already raised his NAD levels with 50-60mg per day. But Vincenzo said that in mice they used the dosages that in human would be 1700-1800mg. So, me too, I hope using in the range of 50 to few hundred milligrams would be of help already. Surely it depends on how effectively our bodies individually use the NAD, so wo still wanna control for inflammation in the body and also for NAPRT activity (I expect it requires mostly similar cofactors and boosters to NAMPT - magnesium, zinc, B vitamins etc).

Can't find any reasonable way to get this in my diet. Highest source is Japanese radish which is only available in Japan. Second highest is coffee, but I refuse to add any amount of caffine to my body even in decaf amounts of 2mg. Fenugreek seeds in high amounts would not be good news, as the interviewee suggests they may have adverse effects.

Fenugreek seeds have 90mg/g, and 18g would get the mouse equivalent dose of 300 mg/kg. Would a lesser amount of the seeds increase NAD via trigonelline? That experiment and video are coming soon!

I don' see fenugreek seeds being harmful for most people in any way tbh - quite the opposite. Actually I can't understand why people would even expect that 😂🤣 - it used for treatments for centuries, made teas out of it etc. - and its part of curry powder I have personally used for ages and I love the taste of fenugreek - been purchasing it in half a kilo bags in the past without knowing it has niacin in it lol BUT I now I should damn increase the dosage of it!!!! - I clearly remember a good cheese that was filled with fenugreek seeds, I quite liked that. Sweet taste kinda. Wonder if I'm gonna start hating the taste when consuming spoonfuls a day.

I use a 1/4 theespoon fenugreek in my soups ..... but the problem is that my smell is going strange and my pee is smelling strange. I was advised 10 years ago to take fenugreek by a Chinese Traditional Acupuncturist .

Nope, TMG = trimethyl-glycine, whereas 1-methyl nicotinic acid = trigonelline are completely different metabolites.

Exercise is a constant in my approach, yet NAD was relatively low without NAD precursors, so there's more to the story than general recommendations. The key is testing, to find out, imo

Sorry, can't bring myself to use that platform anymore. From the abstract, it seems like this might be only a problem with ppl having a particular SNP. I suspect another indicator of a problem might be eczema since niacin supplementation is associated with increased risk of eczema (women-only study), severe eczema is associated with increased CVD risk, and VCAM1 blockade reduced eczema in a mouse model. ~ Would different NAD precursors have different effects on the production of these metabolites? Or would some whole foods have better effects on overall niacin/NAD metabolism than supplements? That's usually the way it goes & I'm guessing that's why you prefer food to supplements. @@conqueragingordietrying1797

PS Vincenzo said (@ 46 min) that 3 to 4 cups of coffee increase plasma trigonelline to about 8 to 10 micromolar and was likely not enough to significantly impact NAD+. My Iollo level was 1.8 micromolar, much less than 8, yet I am a coffee drinker though not right before blood draw. So now I'm wondering whether foods can affect NAD+ via trigonelline, and if not why not. Is this just a freak thing where you need very high levels of trigonelline, outside of dietary ranges, to move NAD+?

The only way to know is to measure NAD-it should be pretty good/youthful, assuming that trigonelline increases it in people.

@@conqueragingordietrying1797do you think fenugreek sprouts would have higher amount, just like broccoli sprouts? It may be easy to add fenugreek sprouts in the salad. There are published studies showing that fenugreek seeds are beneficial in controlling blood glucose, also increase testosterone.
