What is Y2H, its requirements and how it works? An easy and animated video. Y2H is a molecular genetics technique to validate protein-protein interaction through downstream activation of a reporter gene. Yeast two-hybrid is based on reconstitution of a transcription factor (TF) fragments fused with two proteins whose interaction needs to be studied.

there is a mistake in this explanation you said the selective media lacking trp and leucine and plate containing trp and leu. but in diagram you have written selective media lacking trp nd leu. and plate lacking trp and leu. please correct it very confusive

My videos usually are based on multiple resources. For this video I read some research articles and books and covered basic concept. Based on my imagination gave it animated. If you are a research student, I recommend you to refer related research articles.

How do we make sure that some other transcription factor (instead of our plasmid inserted bait & prey) can't turn on the transcription of the reporter gene?

Thats an excellent question. TF binds to specific UAS sequence and not any UAS, hence highly specific in expression of reporter gene. Also, negative controls are always used.

Why can both proteins not be on the same plasmid?

If we place both of the proteins in one plasmid, we also need to place the AD and DBD in the same plasmid. That means the domains (AD and DBD) are already in close proximity, leading to the recruitment of transcription machinery and ultimately results in the expression of the reporter genes. We can,t study the Protein-Protein interaction by placing both the proteins in one plasmid.
