Vincenzo Sorrentino is an Italian-born scientist in the fields of aging, mitochondrial biology and neuromuscular degeneration. He obtained his PhD Cum Laude at the University of Amsterdam on LDL-cholesterol regulation by the ubiquitin-proteasome system. Subsequently, he moved to Lausanne, Switzerland, for his postdoctoral research in Prof. Johan Auwerx’s lab at the EPFL. His work there focused on the understanding of mitochondria and NAD+ metabolism in neurodegeneration and muscle aging. He then obtained a position as Group Leader at the Nestlé Institute of Health Sciences in Lausanne, to lead research focused on integrating basic discoveries on nutraceuticals and their effects on mitochondria and protein homeostasis with their translation into novel clinical applications. Since December 2022, he is an Assistant Professor at the NUS with the Dept. of Biochemistry and the Healthy Longevity TRP, Yong Loo Lin School of Medicine, to continue to develop his research on how metabolism, nutrition and proteostasis are linked and impact on health and aging.

15:13 interesting that this works through the Preiss Handler pathway. NMN & NR use the salvage pathway. It's still an open question, though, whether raising NAD levels provides any health benefit. It seems like it would, but the clinical trials haven't shown much. Maybe they don't know what to look for. Great new format with the live interviews, by the way. Since your guests are from all over the world, it might be hard to maintain.

Hi Paul, thanks for your comment. Why some of the results from mice have not translated into humans is a good point. We discussed it with Dr Joseph Baur, but until we get solid results in clinical trials it is still an open question. Though if the NAD precursor is helping to remove protein aggregates (which we talk about later) it may help sarcopenia and AD but would not be visible in the short time frame of a clinical trial. I would love to do more in person interviews! We were lucky that Dr Sorrentino was visiting Hong Kong so we could meet in person. Hopefully I can get out more this year and have a few of these but most will be remote for the moment.

Hi Abdelilah, thanks for comment. Dr Sorrentino spoke at ARDD about trigonelline and also has a paper based on the research currently in the review process. I will talk about the paper when it comes out.

Another great reason to consume Coffee. Trigonelline should be even contained in decaffeinated coffee. (As a zwitterion it should be barely soluble in organic solvents)

Liquid CO2 behaves as a solvent much like Aceton. As a chemist I would guess that it should leave the Trigonellin untouched while the much less polar Caffeine dissolves.

In the IKON TRIAL COPD is targeted with NR (Nicotinamide Riboside). Is there a good chance that Trigonelline can be used for COPD patients?

One more way to keep NAD+ up and skip all the NMN and NR hype and expense. I recently learned that coffee ( I drink 3 cups of decaf/d) was high in trigonelline (prior presentation by Vincenzo). Also eat other plant foods high in trig. Turns out in testing my blood my trigonelline level (fasting) my level is about 3 times normal (1.5 micromolar per two tests vs about 0.5 micromolar normal plasma level). Haven't correlated to NAD+ level yet however I exercise and do TRE daily which also helps keep NAD+ up among other benefits. Diet and exercise.

Unfortunately many people think truth can only be accessed through prestige, when in reality prestige will more often than not increase the chances that the claims were made with more rigour, because the prestige usually means the person has more knowledge and insight on said topic, meaning the claims have met a higher standard of rigour, than a claim made by someone without that prestige. But what elitists overlook is sometimes prestige can reinforce biases that make truth harder to access. So in short prestige is a very important & vital marker - but only if you understand that it can simultaneously be problematic too.

@@Vgallo I could not agree more. I am a research scientist/mathematician at a prestigious university. My most important works arose when I questioned the status quo and quietly developed my ideas until I knew I had something. However, most research papers in math and science are incremental. Peer reviewing does not catch all mistakes. It is only when others want to cite us that they will go through every detail because their reputations will be at stake. We have a lot to lose if we claim or cite something controversial and it turns out to be wrong. FQXi funds maverick ideas in physics because it can be hard to get funding off the beaten track.

I would add that the key topic Dr Sorrentino talked about and the direction of his study, the translation of aging therapies to the clinic, is one of the major obstacles in longevity at the moment.
