There is one quary I am facing is that when we diregt DNA with 2 types of RE its form 3 types of fragment 1'-1', 1'-2', 2'-2' and if we add 2 types of adapters it will binds to respective sites in all 3 types of fragmnet and we add 2 types of primer for its PCR it will also amplyfy all 3 type of fragment. (its what i have understand). But at many places its written and some professors saying that only 1'-2' fragment will amplify in PCR but 1'-1' and 2'-2' fragment will not amplify and they are putting reason is that forward primer binds to one side of 1'-1' fragment will not bind as reverse primer to other end of same fragmnet which has ligated with same adapter on both side. Please provide me better insight into this confusion. it will be greater help by you. I will be waiting your answer

@@XploreBio that would be great.. looking forward to it. And please do keep an explanation for 1. Whether all these markers are fixed for a particular individual 2. Whether whole genome of an organism is taken for the experiments 3. If the DNA banding pattern is taken into consideration, then why do this dominant and Codominant concept arise.. it's not like we r seeing the expression of the corresponding proteins of the genes... Thank u

No they are universal and not complementary to DNA. You ligate these short adaters to the ends of fragments.

@@XploreBio i actually dont know how aflp is show dominant allele character and rflp as co dominant, because when we use restriction enzyme in rflp is break the dna(as continum lenght) into smaller and shows difference in band of heterozygous allele, but aflp is also used by enzyme to break, how it shows dominant allele mostly

You get huge number of fragments amplified when you perform PCR with preamplification primers because of adapter+1 extra base that confer sepcificity. In final amplification, you ise adapter +3 or more extra base that confer higher specificity for fewer fragments to amplify. This reduces number of fragments amplified finally. It is important for proper scoring of fragments. Too many fragmnets will be difficult to score.

@Pawar. Throughtl pre selective amplification you reduce number of bands amplified and similarly in selective you reduce the number of bands even more so that they can be separated on gel. These 2 steps are basically to have jivh template for amplification. Hope this helps.

Adding those few nucleotides increases specificity to the primers. And therefore fewer fragments gets amplified.

It's like the longer the length of primer, higher will be the specificity. Specially the 3' end nucleotides.

In that radiolabelled probes which becomes visible on photogenic film. If similar probes were used in AFLP, there woyld not ne any need of amplification.
