Complete with clickbait thumbnail! I breakdown scenes from Thymocytes (episode 9) of Cells at Work (Hataraku Saibou) from a real doctor's perspective. I'll be reacting to scenes and attempting to share a little bit more about the science behind the anime!
In this episode we cover the T cell development, the thymus, positive and negative selection, and even have time for a baby photo too.
This is a particularly heavy immunology episode, I've tried to simplify it as best I can, I'll include clarifications in the description here as they crop up... which they will because of the complexity of the subject!

Here is the thing: we all apreciate some healthy self deprecation but mostly it's less about stupid or not stupid and more about explaining (really) complex issues broken down and then connected in a way that makes sense base on your existing knowledge & experiences. That's why doing this in the context of an anime helps and that's what Ed Hope does well. Use examples that we can understand (remember the 'bus' metaphore) and then recombine it in the context of the show.

While it also shows what a difference a good teacher makes this is one of the anoyances of my life. I had such a large span of my life dedicated to learning and I resisted so much and now (that I have a full time job an 'adult timetable') and need to squeeze learning into my day I CRAVE new information and whenever I feel I didn't learn anything new in a day I feel really dissatified. Oh Brain and your ways of working xD

I'm not big on "reaction" videos and it could write up a thorough treatise on why that is _(but I shan't),_ HOWEVER, I was pleased that I made an exception, for this particular video. It's like an extended education, on an already educational animated show, as well confirmation and explanations to what we're seeing; I quite like that! I know "react" is a popular term on RU-vid, but I feel "Confirm" or any other fitting synonym would be more fitting. I have not watched _'Cells at Work'_ myself, even though it looks pretty entertaining, on top of being informative, as is intended. I DID however watch _'Once Upon a Time... Life', (1987~88)_ which was MY generations _'Cells at Work',_ I suppose. Clumsier animation, but still left me with vital information about my body, that I remember to this day. I hope _'Cells at Work'_ will do the same, a whoppin' 30+ years later! *Quick edit:* I just went back to see the first episode of the series, and you DO mention _'Once Upon a Time... Life'._ SPLENDID! 😄

Mr. Al Natural Killer Cells are somewhat separate, although they are also lymphocytes they are part of the innate immune system where the Helper T cells, Cytotoxic T cells and B cells are all part of the adaptive immune system. All come from the same progenitor but NK are sort of separate from the rest

Additional notes: 1. We found out this episode that Helper T Cell was actually a pretty good combatant, kind of like a prodigy. Meanwhile, Killer T C was actually kind of a runt, working hard to just be competitive. The dynamic remind many of Naruto and Sasuke, with Regulatory T Cell I guess representing Sakura. 2. Helper T was actually not that nice towards Killer T, but they were forced to be in the same room. Because of that Helper T finally saw that Killer T is more than meets the eye, and that he is working hard to be the best T Cell. 3. It all showed up when Helper T saw Killer T secretly training that "technique", which because of that, Helper T helped Killer T overcome his fears by reminding him of that "technique" which helped him "graduate" (albeit accidentaly). 4. At the end the two had the animé cliché of punching each other, showing themselves as sort of best friends now, even though they still kinda hate each other's guts hehe.

I love it when you get into the grit of how the processes CAW tries to simplify work. I think the T-cell selection process you laid out was pretty easy to follow, and offered an insight that I frankly didn't get when I watched CAW the first time through. In the show it seemed a bit more 'random' which cells were gonna pass and fail.

I have learnt so much from you and your educational break down of complex systems esp throughout this series. I'm enjoying how much you can see their adult personalities in them. Helper T is always trying to get the most out of Killer T, and during the final exam he provokes an appropriate immune response from Killer T. Meanwhile Regulatory T is forever telling Killer T and Helper T to calm it down and focus on the job. Their professional qualities are their innate qualities which the thymocyte has simply nurtured.

It's mostly the same process, different proteins tested but the concept is the same. Tregs are selected in the negative selection; in the long and short is Tregs are what become of t-cells that don't respond strong enough to trigger the death signal but too strong to become mature naive Effector T's. It's called a Goldilocks process, not too strong, not too weak but just right. That being said, Regulatory T was present in the same bootcamp as Helper T and the Killer T captain- she's with them in most of the training scenes and that's kind of all there is to it, there's not much materially different to show regarding how she differentiated. They all started as the same progenitor cell and specified during this period in the Thymus.

Didn't think the macrophages they showed also played a part. That is indeed nice detail on their part and there was a lot of explanation in this video in particular. The parts about positive and negative selection was very clear.

The thymic macrophages are the antigen presenters actually producing the class 1 and 2 MHC used to put the developing T's through the selection processes, plus you know, eating all the ones that die from failing either test.

The show is based on a comic/manga though, and the author has done a lot of research and has medical advise as well to keep it as true as possible while still keeping it entertaining. I really recommend the original work (which is available in English) too! It's great!

It's very on the nose in a lot of places, not just in how they mechanically work but how they are visually designed (ex. Eosinophil and Basophil's pink and blue uniforms match their Gram stain colors); not quite right or oversimplified in others. Most of the inaccuracies are mainly just conceits required for having persistent characters to follow or to make the information digestible and easier to understand. Examples: the average lifespan of a platelet is about a week, so we would not actually be seeing Leader and Backwards Cap-kun (yes that's his name) as often as we do; similarly, a Neutrophil's lifespan is measured in hours, so U-1146 is not surviving a week long battle against an influenza infection. Slightly more complex example, the Monocyte scenario (coming up in a later episode) is a little oversimplified and omits the connection to Dendritic Cells, but it gets the visual point across about differentiation. So all that being said, the main series author, Akane Shimizu, is very well researched for the series as well as the the now 5 spinoffs she's involved in.
