I sent my dog's DNA to three different DNA testing companies and the results all came back differently! Let's talk about why: reference panels!

Thank you so much for making this video! 😊 This was the BEST explanation of these tests I’ve found yet. I mulled over getting one of these tests done for my new rescue dog but interestingly I am starting to come to the conclusion that I can, with 99.9% certainty and no assistance, declare that she is, in fact, a dog. 🐶

American Staffordshire terrier and American Pitbull is often considered the same exact breed. So I would say those tests where rather consistent with one another.

2 different breeds and are not considered the same. I believe the later of the breeds was just added to one of the register lists. In depth owners will call you out on this to and some get pretty defensive.

These two are separate breeds as I understand, but both are considered pit bulls.

Pitbull is the slang for fighting dogs....American Staffordshire Terriers were not bred for fighting.

Oh man, I can't imagine all those draining hours of painstakingly having to compile so many cute dog pictures. Science communication can be such a cruel and demanding task sometimes. You really took a bullet there for us!

I just used Embark because they were the only one that had Carolina Dog in their breed panel. I can't wait to get the results!

I used wisdom panel it's the most accurate DNA test, I got my results in 2.5weeks I have her family tree and relatives closest relative is a brother from a previous litter!!

2:48 - Y'know how they have the t-shirts with chemical structures for molecules like caffeine? I think you should try to find the part of the DNA sequence for a couple of these traits and make shirts with that sequence and the caption, "toe bean gene" or "boopable snoot gene". That would be some great merch!

I mean, I'm sure there isn't literally a single gene responsible for each of those particular traits, but maybe you could find something vaguely close enough to work for a cute shirt. (Or I guess you could just make up something plausible.) I can imagine a shirt with three or four panels/sections (four or nine would work if it was a grid layout), with a traditional double helix drawing in each (to make it more visually attractive than just a bunch of letters), with the ATCG labels along each. Then each diagram could have one of those labels ("boopable snoot gene") -- either right below that diagram, or with an arrow pointing to one nucleotide (or section). Heck, maybe the nucleotide(s) being pointed out could be little paw prints. Other good ones could be "puppy dog eyes gene", "floopy ears gene", "squirrel chasing gene", "tail wagging gene", or "fuzzy noggin gene" -- or some cuter way of saying each. But I think the last one should be something like "best friend gene".

It’s worth going back and checking later on after more data is added. My dog’s DNA results changed pretty fairly drastically on the wisdom panel. Going from 6 or so breeds up to 10.

I thought you were going to send your dog's DNA to a human DNA tester to see if they would reject it as not human. That might be an interesting question as well. I'm sure they get a tiny fraction of those.

So some people have done that, and they act like it's a big "gotcha" when the company sends back results saying "You're canadian!" when they're actually a chihuahua. But many of these companies just use a chip that's looking for tiny pieces of DNA to bind, and there is a lot of DNA overlap between humans and dogs (we're mammals with similar organs etc etc). So yeah, some of the dog DNA could bind to the chip and give some calls. It won't mean anything (because reference panel selection is important!) but it's like getting back puzzle pieces with the right shapes, but from the wrong puzzle. You could fit them into the missing spots on your "human" puzzle and they wouldn't make the right picture but you could still squish them in and return a result.

I started to realize this when I tested my puppies. Both my girls are litter mates. Interesting thing was, one of my girls came back with one extra last percentage being golden retriever. While the other stayed at 5 with the last being lab. I also found there brother’s results as-well through relatives. And he was generally the same EXCEPT he had the last small percentage of super mutt. Estimated being Akita inu and cocker spaniel. Which I didn’t see on neither of my girls results. I tested with embark. It was interesting.

I'd love my dogs the same if they were part loch Ness monster, because I feel like maybe they can be worked with. Now Cthulhu, I'm not sure. Does not seem eminently trainable... :). In all seriousness I am so chuffed that I found this video. My husband and I were early adopters of 23andMe, so I'm very happy to hear a scientist's explanation of these tests/services! ♥️

Nice video very educational. I just sent my dog’s dna sample. Just a little worried cause i could not take the sample for more than 5 secs each time. Lol. Hopefully i got enough cells.

Great video! It’s nice to hear from a dog owner who has deep knowledge of genetic testing. I get why they ask your breed but we submitted to Embark without giving them anything more than “mixed breed” and our dog’s age. In reality our dog is a “designer breed” with two AKC registered purebred parents. I kind of expect Embark to mess it up for the reasons you mentioned.

A question I've never gotten answered: Everbody always says DNA are the instructions to build cells/body, but what mechanism carries out those instructions ? Instructions by themselves don't do anything. what mechanism takes raw materials and puts them together to build a cell and eventually a body ?

This was super informative! Thank you! Is it beneficial to call one of the companies to see if they have reference samples for the Catahoula Leopard Dog breed? My dog is mixed & supposedly his mother was Catahoula.

I used Wisdom Panel, because I suspected my dog was part Catahoula Leopard, one of her eyes is multicolored, half blue. Found out she’s 75%Am Staff, 12.5% Boxer, and 12.5% Australian Cattle Dog. Now I completely understand her habits, mannerisms, and behaviors and can see each breed’s influence. Pretty cool, I knew her mama was a Red Nosed Pittie, but wasn’t sure about Daddy, he looked like a Pit mixed with maybe Weimereiner. I’ve had other dogs tested, where they came back with 15 or more breeds, which isn’t very helpful and likely not very accurate. I’m not sure if Catahoula is one of the breeds they test for, but I would think it was. Now I’m curious.

This was a great video. Our dog’s DNA came back as 100% beagle from Wisdom Panel. However he certainly appears to be a mix. Should I question their 100% certainty?

What fascinates me is the tremendous variation between dogs that is seemingly unmatched by any other pet bred in similar circumstances. I did look in to it once but found a few explanations ranging from: It being predominantly down to the length of time weve been breeding them. To: There being more fundamental genetic reasons for the changes (At least one paper calling it a 'slippery' genome). Ive learnt from similar questions that the answer has a tendency to be somewhere in between, or perhaps the former led to the latter, its still quite opaque to me.

They do have an absolutely remarkable amount of variation. Looking at a dachshund and a german shepherd and remembering that they're the same species is kind of mind blowing!

With Embark it’s more than just breed identification they have over 140 known genetic diseases. Many of which if you know you can adjust and have your dog live a longer healthier life. Also just cool knowing the history of your dog if it’s a mix breed.

Have you tried the apps on your phone that tries to identify your dog's breed just from a single photo? And see if they match with the DNA results you got?

@@AlexDainisPhD My dog is a purebred miniature pinscher, which is not very common and the app was able to identify her as 100% min pin. I also tried it on my mum's dog who is a mixed breed and it seemed to pick the right mix.

What three companies did you use? I only heard of one but didn’t notice the other two. I’m curious if the others because I have a dog that isn’t quite est terrier (her coat is too soft to be a real rat terrier) but she is definitely rat in appearance and temperament, but she’s so old that when she passes I would like to find one like her to keep me company . Thanks for the help

As soon as you said you were told that she was part corgi, I went 'nope'. Now don't get me wrong, I don't know a lot about dog breeds, but there is one thing I know for sure about corgis. Corgis have two copies of either a dominant or co-dominant (can't remember which) dwarfism gene, which is why their legs are stubby. So all corgi crosses look like a corgi that's cosplaying as another breed, because even when they inherit just a single copy of this dwarfism gene it will still be expressed.

Would have been nice to compare the results of inherited condition markers...I just sent my mixed breed puppy's in to make sure she's not particularly prone to DCM, since I head she has some dobie in her. We'll see what Embark says.

Thank you so much for breaking this down. ❤️ I was so upset with the results we got for some of our dogs. One was that our dog results came back as 100% American Bully and they gave us a list of relatives. However at least one of those relatives had 0% American Bully. So how can they be related?

Pretty consistent! Mainly Chihuahua, Cocker Spaniel, & Pitbull (which I felt in my bones she was! She has the squared/strong cheek and jaw arch of them, that's how I knew mine was part Pit before her DNA test confirmed it!). Looks like 1/3-ish Chihuahua, 1/4 Cocker Spaniel, & 1/8 Pit (which is often a mix of Staffy or AmPit).

same here.....my little [30lbs] rescue has crazy DNA.....at least i know why she pushes my Rhodesian and pitt /malinois around.....she's a big dog in a little body.....big bravery.....they are cool with it & are good to her !..........she's pitt, rottie, GSD, ACD, & other breeds in trace amts....

At the beginning of the discussion about Penny, I saw her running down the hall, and I immediately thought of my sweet Heidi, who I lost on 1/19/2021. And then I thought, why do I see Heidi in Penny. Heidi's little front legs were no longer than 2 inches long. She had long reddish hair along with a floofy tail. But that sweet little face on Penny just shouted out dachshund. I thought I was crazy. But then I saw that one of the tests said she had dachshund. Also, judging by the way she behaved on the video, she seemed so much like my baby. Then when you said she liked chicken, I thought that's it , they are distant relatives. BTW, that's her little pic on my comment.

I wanted to find out what other breed is in my dog other than Pomeranian. Her hair is not double coated which most Poms are. I bought a kit called DNA my dog. I unwrapped the plastic and when I opened it I only found a box inside. Nothing else was in there. I have never done this before but I know there was at least supposed to be swabs. I contacted the company "Petsmart" and told them I found things were missing from the kit, so I have to take it back and get another one. I will this time get them to open the kit so we can make sure what was supposed to be in there. Kind of disappointing.

Question: Were these DNA tests double blind? I ask because many of these companies give the option to include photos, videos, and descriptions of the dog. As a fellow scientist, i think that's super sus...

Given how these DNA companies (both human and dog) work, I doubt it. They literally just put the processed sample into a chip, feed the output from the chip into the current version of their statistical model, and send you the results. There's not really any room for fudging, since they will also give you the raw data from the chip at your request, which would allow anyone to refute any nonsensical data, destroying their reputation.

The interesting thing is that whether you become a dog owner or not might also be determined by genes. At least that's what recent study says. See "Evidence of large genetic influences on dog ownership in the Swedish Twin Registry has implications for understanding domestication and health associations".

I clicked here because I just got the results back from Embark and I am pretty sure they are not correct. My dog looks (and behaves) like a certain giant breed (ancient) and does not look (or behave) like the DNA results I got at all. I am now curious about trying two other companies just to see. EDIT: I think you explained why the results are so off.

Now all this info made me wonder what type of dog I'd come out as if I took the test instead of my dog... 🤔🤭 Especially because my husband thinks I'm a Tazmanian Devil...

After seeing Penny, I'm going to guess before I watch the rest of the video. I would guess Chihuahua and Beagle. Maybe a little Jack Russell. (edit: I wouldn't have guessed THAT!)

@@AlexDainisPhD Certain breeds seem to mask the traits of others in mixed parentage dogs. Chihuahua is usually dominant in a cross, and we've all seen dogs where you know one parent was a Black Lab, but who knows what the other one was.

I don't see the Great Pyreneese :-). I would have said jack russell x cavalier or beagle but chihuahua x cocker seems quite accurate.

Your paternal great grandparents were pretty much 100% Lithuanian so that 25% is pretty much spot on! (Don't be weirded out, I'm your second cousin..) Cute dog! We have what we believe to be a Jack Russell Chihuahua (rescue dog) and she looks very much like your Penny just a little smaller and black instead of brown. We should probably have her tested to find out for sure, but her aggressive and very hyper behavior tell me we probably have it nailed.

I mentioned this in another comment, but some people have done that and it's kinda unfair. From my other comment: "many of these companies just use a chip that's looking for tiny pieces of DNA to bind, and there is a lot of DNA overlap between humans and dogs (we're mammals with similar organs etc etc). So yeah, some of the dog DNA could bind to the chip and give some calls. It won't mean anything (because reference panel selection is important!) but it's like getting back puzzle pieces with the right shapes, but from the wrong puzzle. You could fit them into the missing spots on your "human" puzzle and they wouldn't make the right picture but you could still squish them in and return a result."

They should *probably* throw out the sample for being low quality, but if you get enough calls... I can see why sometimes they might return results.
