We can use a small tag in the protein and express it in the cell..let's say you want to localize it to the nucleus then you can use a nls signal...

FISH is a broad technique used for detecting specific mRNA ..... serotonin bindings receptors ot 5 HTR mRNA can also be detected by FISH. It all depends on what you wanna ask...so expression of 5HT mRNA can be monitored in spatial temporal fashion but the receptor functionality can’t be studied with this technique

There is difference between isolate and detect, hybridization can derect but cant isolate it for further downstream use
