Acclimating tissue culture plants can be a challenge especially if it’s new to you. I’m showing every step that I take to have 100% success acclimating any plant from TC!

I'm waiting on my very first TC and I'll be honest, I had no idea what I was getting into. I just happened to look up the term tc+prop tonight and seriously I have been through so many videos & felt so confused and overwhelmed until I got to your content. Very informative and straight forward and detailed info. You're a lifesaver. I can't wait for my new little dude to show up!

I’m so glad this was helpful!! You got this 💪 it’s intimidating dealing with your first tc plants but once you get the hang of it, it’s a ton of fun watching teeny tiny plants grow into their full size

I haven't seen your content else where, but you've nailed this vid. I'd never heard of that media. It takes a little longer, but I would love to see a comparison of different methods or protocols if you do Tc yourself. Great job! Looking forward to more content

The only thing I was not clear on was, when using two plastic cups as a dome, did the bottom cup need drainage holes?

@@vivity01 nope! I keep a reservoir of water in the bottom with this mix so no drainage holes are necessary

Just found your channel and subbed! I have been growing fungi for 27 years and recently started a mushroom farm. I have many industrial size pieces of equipment like my autoclave and flow hood, I have ALWAYS wanted to get into tissue culturing and I think it will be my fall project. The Biocouplers intrigue me. Have you made a video about the pros and cons of using them versus older tried and true techs?

This is helpful. Most of the plants I want are carnivorous plants and the cheaper ones are in tissue culture.

I never used to but I am moving to sending this mix instead! My holy grail used to be coco coir and perlite which is still a great simple substrate to use, but I’ve had much better success with stratum 😃

Please helllp! 😭 I messed up! I accidentally added 30gr/L of agar instead of 6gr/L. I used an expensive DKW media and other expensive additives and hormones and I don't wanna waste it! Will this too high concentration of agar affects plant growth?

🥺 hate to break bad news, but I think so..I imagine the agar will be rock solid from 30g/L so I think it will be difficult for the plant to make good contact with the media. Keep me updated how it goes though, I am curious if the plants will still be able to absorb the nutrients with such a dense media!

Could you use just perlite? A bag of fluval is literaly the same price I paid for my TC plant.

Yes absolutely! I like adding fluval because it releases nutrients but straight perlite with a small reservoir works great as well

I notice your distilled water is PC brand, if your in Canada, where do you buy TC plants to ship into Canada?

TC 101 group is where I've found a few sellers! I don't really buy them anymore, there's one on Etsy as well who I used when I first started and they were great, I think the name of the shop is Plantsme or Plantsme2 something along the lines of that

I prefer without a heat mat, it can get REALLY warm in the sealed environment and speed up potential mold growth or melt the plant from the heat in my experience

What would I do with Aquarium plants , sold as tissue culture , that I want submerged? Supposedly it's routine for them to melt.

I have a few TC plants in my aquarium, they can be a bit harder to establish, i probably lost 1/4 of my dwarf hairgrass when I planted it out so I’m definitely not an expert on the aquatic side of things! The biggest tip I can give is make sure 100% of the gel is rinsed off before planting them in your tank and give it a dose of fertilizer to start growing roots right away

A lot I've kept from past TC orders or purchased from other local sellers who had unused jars! I have found them on Aliexpress too though and they're kind of expensive on there but if you don't need a lot it works great. I believe ULINE also has a few sized jars with polypropylene lids but I haven't tried those yet

To avoid having contamination issues when using working with growth media for plants i have found that the following process is essential after 10 years working with GM plants. *wash your hands and lower arms thoroughly with soap ( most important step) * wipe down flow hood with 70% ethanol. * Before you put your containers, bottles, plates etc in the working area wipe them down with 70% ethanol. * spray a little 70% ethanol on your hands before you start. * Have a bottle of 70% ethanol in your working space ( This is kept specifically for flow hood) and decant some into a beaker to sterilize tools whilst working. * Have a flame ( away from the ethanol) so that every tool is dipped in the ethanol and then passed through the flame before the tool is used. * If working with plates , only raise the lid the minimum and never place the lids upside down in the flow hood.

@@Parkfolia I only use a bleach Solution for seed sterilisation. To save yourself a lot of headaches doing this stuff I'd recommend that people find a published protocol and follow it. For example, this paper (1985). A Simple and General Method for Transferring Genes into Plants. , 227(4691), 1229-1231. doi:10.1126/science.227.4691.1229 Is the most common plant cell culture transformation method used. You can download the full paper by visiting sci-hub This site allows to download any scientific paper even if it is behind a paywall.

If you are working with any hemp, marijuana, cannabis plant this will not work. A virus has been spreading through growers crops called HLVd. Or hops latent viroid disease. Alcohol will not kill it and you will spread it through your utensil's. The only thing that kills it is bleach , I suggest keeping your surface area clean with bleach and your tools with bleach then wipe they areas down after using the bleach with alcohol , your plants will not like the household bleach at 20 percent ,

I use a 20 percent bleach, new single edge razor blades for each plant , then replace it. Then after using bleach I clean the surface areas with alcohol after that. The plants will not like the 20 percent household bleach. Not dichlor, but household bleach.

Keeping a sterile environment isn’t much of a concern anymore once they’re planted, the idea is to keep high humidity and with the cups taped together it keeps the environment sealed at 100% humidity :)

It seems like all the tc videos I see are generic. Mix this stuff up. Throw a piece of plant in there. 2 weeks later you have 1 million plants in a test tube. Hmmm. Ok in a test tube what is considered a plant? 1 root. 1 cell. ??? How to divide the million plants in the test tube without injuring or turning them into a combobulated mess.etc
