Mammalian target of rapamycin (mTOR) was named because a drug called rapamycin inhibited a protein that was then named, target of rapamycin (TOR). The “m” is because it’s in mammals (zebra fish TOR is, similarly, zTOR).
mTOR is central to the opposing biological processes of growth and longevity. mTOR is thought to promote growth and decrease longevity.
Basically, any process you can think of relating to muscular growth and longevity probably has some relationship to mTOR. Here are some examples:
Exercise activates mTOR signaling in skeletal muscles (growth)
mTOR inhibits autophagy (longevity)
Insulin activates mTOR (growth)
Sirtuin proteins inhibit mTOR (longevity)
BCAAs activate mTOR (growth)
Fasting inhibits mTOR (anti-growth and longevity)
And, as precited by its pathetic name, rapamycin increases (longevity), at least in mice
How exactly does mTOR impact all these processes? Well, it’s kind complicated [show image below] and there are actually two different mTOR complexes (mTOR1 and mTOR2) that have slightly different functions. So, for the sake of simplicity let’s just say this: mTOR is a kinase. This means that mTOR acts to regulate the activities of other proteins by adding a phosphate group from ATP to them. Depending on what protein it adds the phosphate to, and where on that protein it does so, is can either activate of inhibit its target and pathways downstream.
The message, in its simplest form, is that mTOR activity makes you live hot (and strong) but die young. But since when is science ever simple? What annoys me is that people tend to be scared of anything that activates mTOR because they are afraid it’s going to kill them. This is problematic thinking for two reasons:
(1) Biology is all about balanced cycles. You need periods of growth (anabolism) and breakdown (catabolism). If you’re all anabolism (like a body builder) you will die young. However, if you’re all catabolism, you’ll die even younger. In fact, one of the leading risk factors for death in the elderly is low muscle mass. You need a functional level of muscular power to live and thrive. No mTOR and you’d wither away. In short, you do want to activate mTOR on occasion. Just don’t overdo it. “Extremes are easy, the challenge is balance.” “
(2) The inside of your body is NOT a homogenous blob! Why do people always forget that fact? Just because you activate mTOR in once place doesn’t mean you activate it everywhere. For example, when you work out you activate mTOR in your skeletal muscle but might actually decrease mTOR activity in your brain. Thus, it’s possible, and I would argue probable (such studies can’t be done in humans without cutting into their brains), that exercise activates muscular mTOR but inhibits brain mTOR, giving you the exact organ-specific balance of activity that you want!

I love how simple you explain stuff! Thanks, Thomas! Keto carnivore here for 1 year, it saved my life. Lab tests are great, eating 1 or 2 times a day, never done fasts and did up to 135h only water, salt, mag and potassium, amazing.

Wonderful explanation on how to benefit between ‘Mtor on’ and ‘Autophagy on’ cycles. I started the Ramadan fasting of 30 days where one goes for about 14 hours of dry fasting everyday. I am going to alternate between foods that promote autophagy when beginning the fast before sunrise and foods that promote Mtor activity when breaking the fast at sunset with some exercise just before that. I think this will be a great strategy to balance these two states on a daily basis.

Well rounded presentation. I have been looking into the relationship between mTOR and longevity (or the lack thereof). New research suggests over activation of mTOR pathways may be a key area in shorter life span. If you make another video on this subject, I will watch!

very interested in mtor and balancing muscle growth with longevity. the question i'm asking is should i maintain high protein intake all the time or should i cycle it. I'm especially interested in cycling of autophagy days/weeks (e.g. fasting, metformin) and body building days/weeks (+ high protein, bcaa intake).

I appreciate your in-depth explanation and would love to understand this better. I've listened to Dr. Gabrielle Lyon talk about this in relationship to the importance of getting enough protein - and at regular intervals. It sort of makes sense but I'd like to understand it better because it seems like it would be a real game-changer. Thank you Thomas - as always!

I would be very interested in a more detailed breakdown of mTOR. I'm a layman (no training in health sciences) but I'm really interested in this. I'm especially interested in practical recommendations of how to build a routine that achieves the balance you're referring to. SiimLand has some videos offering sample workout/diet routines designed to achieve this but I'm looking for more sources. This video was helpful and a good primer on the topic, thanks Thomas.

Firstly, love your channel and thank you for bringing all that knowledge to us, and in a clear to understand language. Would love a more detailed video since mTOR is something I'm actively conscious of and think about (I do resistance training and alternate fasting). Anyhow, I thought mTOR gets turned on when consuming at least 2g of leucine (and with exercise you just stimulate protein synthesis and that's all, so a new thing learned here). I even count my leucine intake when, for instance, I don't want mTOR activated (but consume at least 30g of protein with each meal). So after seeing this video, I admit I am a bit lost with this mTOR thing ... Thank you in advance, and once again hugely appreciate your channel!

Another GREAT video, THANK YOU for all that you do especially sharing your knowledge and expertise. I do have a question if you don't mind in regards to Intermittent Fasting......Technically speaking when can your time start after you have finished eating your last bite of the day? In others words can your fasting window start immediately after your last meal or should it start like 30 minutes later an hour later? Stupid question I know, but I'm sort of OCD and anal when it come to exact time windows for starting and ending fasting periods. Thank you for your help.

Thank you for making a video on mtor. So considering mtor is activated during weight lifting, should I always consider breaking a fast right after my workout instead of breaking a fast 6 hours later so that I can reduce the number of times mtor is being activated throughout the day? I prefer morning fasted workout and not eat anything after that and then break fast at lunch and have a dinner and so on. As I expressed my concern above, I want to reduce the number of times mtor is being activated throughout the day but at the same time want to gain more benefits by combining a better routine. Also if you can clarify if I really have to have a protein shake or meal after a workout opposed to going faster for 5-6 hours after working out. I'm a big fan of your research and clarity of the content you provide. Thank you for everything.

This is the best MTOR strategy that I've heard. While I'm vegan, I supplement with BCAA and additional Leucine before and after a workout.

finally an explanation. ive heard so much about mtor is good for muscles and strength is great for longevity because falling badly when youre over 65 has quite a high lethality rate and then i heard rapamycin inhibits mtor thats why its great for longevity and i was very confused. it sounds a lot like cortison the way you explain it. cortison is important in stressful situations like a workout and afterwards for recovery but you dont want chronically high cortison levels because that has a bunch of side effects that kill a lot of people in their 50s or 60s already

Cold brew coffee with a tiny bit of heavy whipping cream and pyure sweetener is my fasting vice. I always wondered if the teaspoon of cream here and there messed up my mTOR/autophagy. There really isn't much information out there, so I appreciate these videos. Thanks Thomas! Keep them coming.

Ok 👍 so how do we ascertain our sweet spot? I do lift heavy every day... I do IF 16/8 or 18/6 & I do eat a fair amount of protein .... how do I know if all of this is too much?!

Make sure you get Leucine, wouldn't go over 5 grams it can easily cause hyperammonia past 2.5 grams if you go to 5 grams you might want to consider taking zinc and iron or eating oysters to deal with getting rid of the ammonia

Hell yea, this is exactly the stuff i wanna do more research on... been self expirimenting through keto IF for the past several years and i can say from a medatative perspective you can definatly feel the difference in consciousness between the two states and which one your bodys leaning more tword... what I'm super intrested in finding out is the ratio between the two to acheive ones optimal balence over the function of time... like you said its not a switch and, both processes are usually happening simultaneously one more so than the other but hypothetically if from a balanced state i spent 24hours in 100% mtor building mode, then would 24hours of 100% autophagy recycle mode restore the balance? I suppose this is an impossible question as everything is individualized and situational but don't you think it'd be cool to have some sort of device, kinda like hoe a glucometer works as to tell you which state youre in and how much so?

Is it good to break a fast with sardines in brine? I know it's got a bit of fat in it but also high in omega 3 which makes me think it might be better than a protien shake for example

How much time would it take to activet mTOR? On some videos you mention that after a breakfast a 24h fasting finished with training and bcaa+creatine+carnitine and lean protein would trigger mTOR. Would this be a 15minutes or even hours?
