The PPi that is released from apyrase when no light is detected, does that react with sulfurylase which converts it to ATP? and do we still see light? (Apologies for bad English)
I am not 100% sure but I believe Apyrase releases 2Pi molecules instead of a single PPi molecule when degrading the dNTPs in a well. These Pi molecules are not sufficient for the sulphyrase reaction that makes ATP. Subsequently, the reaction with luciferase does not occur since ATP has not been made.
