Take a sterilized lid from a clean Mason jar. Place cap on lid. Add one drop of rainwater. Cover lid with jar for 24 hrs. Carefully remove cap. Use lid to top a jar with water and corn syrup. Colonization!! After a week suck up with syringe.

I have been loving your channel, thank you. I have done strain isolation on petri dishes, and although it worked it was slow and tedious. This seems like a much quicker way at getting results.

Love the work you guys are doing, after learning about the health benefits of mushrooms i began growing pleurotis, hericium, lentinus/lentinula, agrocybe/clitocybe....... Depending on which academic ur talking to and the date of the description. So much with names is changing lately. I learned most of my basic lab work from your videos so i have to credit all ive done since to you. Im now preserving indiana wild species and also breeding my own mushrooms now using mon-mon and di-mon pairings. Thank you because of this video i learned to take cleaner prints about a year ago and my whole game changed. Again thank you

Better idea than cooling your flamed scalpel with the water: wait for it to cool down to the point that it is no longer visibly red hot (only takes 3 or 5 seconds) and hit it with an alcohol swab for a few seconds. Then hit it with a fresh paper towel and she's ready. Then dip it in the water for the moisture if you need. This is the proper way to do it actually. Awesome video btw! I'm gunna check out your channel.

I've tried getting spore prints off of shittake and chantarelles and always get contamination. Next time I'll try the chopsticks thing. I love it.

This stick between the mushroom and the foil is a really good idea, altough I will go for metal rods that can be disinfected well, to really rule out contamination.

Zit nu nog in de fase van uitzetten in graan en van daaruit verder. Ben alleen nog op zoek naar de juiste samenstelling van het groeiblok. Ik lever mondjesmaat aan restaurants. Deze video's zijn heel waardevol om kennis te verzamelen. Zeker als ze zo open en eerlijk worden gepresenteerd.

I just wanted to thank you for the informative video, so many that are giving good info for first time growers are by and for people growing P mushrooms. I am sure I am on a list now thanks to that... I do have one question, even though I see lots of people doing it with no issue, I opted for a inductive heater to heat up my instruments, is that better, worse, or the same as actual flame sterilization?

I so look forward to your videos! I don't know why you would skip the agar step where a person can at least try and visualize and isolate bacteria/mold/etc. Why jump straight to a bag? "incompatible strains" with lines of demarcation on the agar - in the same species? Is this possible? Might it be contams instead? I fruit the mushrooms in the sterile grow bag, rinse/wipe the bag super clean, open in a flowhood, stick a gill piece to a petri lid with petroleum jelly and place over the media filled petri dish, wait a few hours, and replace with a new lid. Voila, sterile spores. I then plate out what I think are sections/isolates visually on agar as they expand. If you haven't tried my "stick a sterile gill to a lid" trick, give it a whirl, and report back.

I've done that before and I do like it very much. Truth is, it's just so little labor to do it with the syringe and I'm getting good results from it so I tend to go with the lazy way there! Lol.

Great video. I notice in the no fuss method you appear to be using a hepa filter negative air system. Is that to avoid spores flying around? How much danger is there? What were to happen if mushroom spores flew around a room? Thanks

I remember commenting about how wild mushrooms can grow in spite of billions different types of microscopic organisms. And that it may have to do with developed immunity, and how exposure can increase immunity of organisms. We know this because of how vaccines work, introduction of weakened strains of viruses. And the fact that kids that play outdoors are known to have stronger immunities than kids that don't. And culture immunity, example Europeans don't react as badly to pox viruses as African or native Americans due to the fact Europeans were exposed to it far longer.... But what's interesting is genetic diversity. It's noted that species with the greatest genetic diversity have the most immunity to not only pathogens but changing environments simply because they have a greater Gene variation to tap into. If one parent is white, the other black, your able to use genes from both parents and their ancestors exposure to protect urself than a child with both white parents. So I wonder if that's why cultivated mushrooms succumb to disease easier than wild mushrooms, their Gene pool will be much smaller. At zoos they have a trade program to ensure captive wild animals maintain as much genetic diversity as possible. I wonder if a trade program amongst mushrooms, mixing spores of same species from different locations and wild versions could strengthen the cultivation?

I routinely bring wild mushrooms into cultivation and they have the same problem. It's really about how fertile the substrate is. Our rich substrates that support high yields are way richer than mushrooms would typically grow on in the forest. Therefore speedier organisms like contaminates are able to colonize your substrate quicker than your chosen mushroom species.

Do you inject lots of spores to speed up colonization, and production of mushrooms tolerating mild contamination because the tissue sample from the centre of the mushroom is pure? Or do you throw away grain that has contaminants?

Great video! Working on a wine cap bed in FL. I’m not a mycologist or biologist, but could I (in theory) potentially find a recessive gene for a heat-tolerant strain? There is a FL Blue Oyster strain on the market.

The cheapest way to make an injection port is just to buy a tube of high temp (red) silicone adhesive and squirt a glob on the outside of the bag lol
