Could I do this same technique with popcorn tek? I always see vids with people using agar, but never inject the spores directly using popcorn tek?Cheers.

i’d recommend using 70% isopropyl alcohol instead of 91% bc the 91% evaporates too quickly you’ll get it much more sanitary with 70%

If you keep your liquid culture in the refrigerator can you add it to the grain while it is still cold? Thank you in advance for your answer. I keep mine in the refrigerator.

Plz guys your needle tip does not need to glow orange when sterilizing just a quick 3-5 once over with the flame then a wipe with the alc is plenty. When it glows orange you can introduce new contams threw flame soot.

What size particulates (micron size) will tyvek filter out? I wonder if its micron rating is small enough that it could be used to build a homade laminar flow filter.

I have no idea. And given that filtering is not its primary purpose, I'm not sure you can find reliable numbers. BUT, I say give it a try for your DIY flow filter! :)

question, what about the filter tape over the hole for the gas to evaporate out ??..i have seen others use the tape .

Mycelium at this stage need very little fresh air. These jars are not airtight and the small amount of air exchange that happens is sufficient. Filter tape is not necessary.

Do you have any more advice on the environment the jar should go in? ie: Light, No light, some light, humid, not humid, really humid. room tempature (Which room) my basement or attic (Sorry, auto correct spelled attack)? So 65 degrees or 90 degrees? Thanks in advance.

I was taught to keep them at room temperature and cover so it's in the dark. I think that is the ideal. But I've grown them in quite a temperature range, but mycelium grows slower at either temp extreme. I had really good results covering them in one of those mylar "space blankets." That was a warm environment with some light (as the mylar is slightly see through).

hello, will the spore spawn directly into the mushroom substrate instead of grain?

Theoretically yes. But applying spores directly to a bulk substrate will usually end in failure. For success spores should be added to a sterilized media (grain jar) so healthy mycelium can be produced in a clean environment free of contamination. This way you have lots of healthy mycelium to colonize the bulk substrate and get a head start on any competing entities (mold and other contaminants)

I bought pint size for my grains instead. I am guessing 1cc to 2cc? Is this going to mess me up?

Pint is actually the ideal size! Pint is the size I am using in the video. (If you got smaller than that, you may have half-pint). And yes 1cc or 2cc is more than enough depending on the spore concentration of your syringe. More than that and you may add too much moisture with invite mold growth and inhibits mycelium growth.

After the mycelium fills the jar, how long would the jar last without opening it?

It depends on a lot of factors, but I'd use it as soon as possible. It will begin to wither, dry up, and die in a month.

Depends on the concentration of your syringes. 0.5 to 1cc will probably be sufficient. It doesn't take much with well done syringes.

What about the other side of the filter, did you sterilize that before closing the jar?

The whole thing is sterilized in a pressure cooker as a single unit. See my previous videos on the topic, this was one part of a long series.

+Nick Woska Just to clarify, I wipe the top with alcohol just to wipe away anything that may have settled on top so I don't push those contaminates into the jar. The underside of the filter paper is still sterile from the pressure cooking in the previous step.

The difference between doing science and just fucking around is writing it down ;D also, lulz, just the tip... 😂 In the case of potentially using silicone instead of tyvek, do you think it would be necessary still to cover the puncture, or would the silicone be self-healing enough to snap back since the gauge is pretty thin, as on a repuncturable medication bottle? Also, how do you think it would work using the sticky part of a waterproof bandaid and peel-applying it, since it was in a sterile environment inside of the packaging?

+Chelsea Tarver LOL! You are right about the silicone being self-eahling enough, especially with a very thin needle. I think that should be fine. I don't know about the band-aid, it's worth a shot! I couldn't say for sure.

Depends having on many factors including environment, substrate and innoculant used. Anywhere from a week to over a month.

Can I heat up the tip, then wipe it with an alcohol paper towel, then inject it?

That's what I do and have not had an issue. Some people have the opinion that you should not wipe with alcohol after heating and some say you shouldn't heat at all and should only use alcohol. But I have done both and been fine.

In the fb groups they tell everyone to only inoculate grain with agar. Does it get contaminated often this way? I mean they preach this.

I think certain techniques just get trendy. Agar, liquid culture, and spore syringes to make grain spawn are all valid options. Each have their pros and cons.

A coffee filter will not likely survive the heat / moisture of the pressure cooker sterilization.

Where should I keep the jars after inoculation? I also got a few pieces of vermiculite in my spore syringe. Am I able to save my syringe with a few pieces of verm in or should I just toss it? I prob have a couple of ccs left.

+HotDommy Keep them someplace dark and warm. Ideally the tip of the syringe should not touch anything while innoculating. But honestly it's probably fine and I would keep it. Just mark whatever jar you next impregnate with that syringe and note that this jar might have a greater chance of contamination.

@@punkyroo right now I put the jars in a closed plastic container by a vent so they can stay warm. It's my first time so I'm really nervous. I also didn't wipe the injection sites on a couple of my jars. The instructions I have aren't really that clear. But I decided to wipe the rest of tops. I hope I can atleast get 1 successful jar lol. I'm just worried because I didn't sterilize the top that they will be contaminated. 🤪

You usually wipe the tops with alcohol before you inject and the cover the tiny injection hole with a bit of clear tape. And keeping them in a plastic container is probably fine. I usually stack mine in a big cardboard box.

I have two holes in my jars. One for my syringe and another hole for air. This jar doesn't have a hole for air. I thought live tissue needed air to grow.

The tyvek has microscopic holes in it as well, the jars are otherwise not 100% airtight. The growing mycelium gets all the fresh air it needs. Extra air holes will only invite mold spores into your jars.

Cheap birdseed with the sunflower seeds removed. So mostly millet and sorghum.

+Jesper Hellvik The jars are not 100% aittoght containers. For mycelium growth, you don't really need fresh air exchange like you do with the fruiting stage.

Punky Rooster I always use lids with holes covered with micropore tape, even with the BRF tek. That’s just what I have learned. Those jars SHOULD be airtight to avoid contams, but still be able to breathe. Free air exchange (FAE) is what is needed during the fruiting stage, but as far as I know gas exchange (GE) is important for the colonization stage...or at least it speeds up the process and it helps to prevent the mycelium from stalling. Again this is just what I have learned from loads of videos and tutorials.

The jars are tightly sealed, but I am making the point that no jar is going to be 100% hermetically sealed. Gas can still exchange slightly. And I've done it this way for years now with great success, so mycelium doesn't need any more air exchange than the very slight amount that happens in a jar like this.

Very little in the grain spawn phase of growth. Fresh air is more important to fruiting. These jars are not 100% air tight and the small amount of gas that does exchange is sufficient.

@@legalizekansas1398 no, you only use about 2cc per jar. However if you inoculate one jar, you can use that jar to make 10 more jars. There are many videos on RU-vid that show you how to cultivate mushrooms. The shrooms youre wanting grow the same way. So once you understand the basics its gravy after that...
