Instant Pot to Sterilize Liquid Culture

Liquid culture is AMAZING. And it's time I tried to use my Instant Pot to make my own liquid culture jars. The Instant Pot has proved successful in making grain spawn jars as well as pasteurizing bulk substrate. In this experiment I try distilled water, rain water, and filtered tap water to see if there is any differences. The recipes is very simple (Thanks Kim!): 1 tablespoon of Karo light syrup, 1/4 cup of coconut water, and then fill the rest of the pint jar with clean water (of your choice). I then put them in the Instant Pot for 30 minutes at high heat. The color changed a little in the Liquid Culture solution, so some caramelization may have happened. We will wait and see

Glad you are giving it a go! Once you have your own supply of LC you won't look back! Bummer about the distilled cracking, your approach to experimentation with different materials is always fun to follow up on. The light golden color is totally normal and fine. You shouldn't have any trouble with those due to caramelization. Contamination is the biggest threat, so long as the sterilization went well and the injection was clean, you should be on easy street. Can't wait to see your results!🙌😍

Nice little video. I have a pressure cooker that can get to 15 psi and my first batch was dark like yours (15 psi for 30 minutes). I think I'll try 15 next time.

Use boiling water before pressure cooking. It makes it less time required to sterilize and more sterile

I did this using tap water filtered with a Burkey . The first time I went 20min and they turned yellow. Next i tried only 12 min and they came out clear. Used high pressure cook setting , no cooldown . I inoculated one with morel spore and it grew very well. On another note, the grain jars i set up with the instapot are doing great except for 1 that i tried using birdseed and got some green mold. The seed may require longer time in the pot. The others were brown rice. Those were done for 90 min.

Only 12mins? Is that enough? I also have seen yellow ones just do fine. As caramelization of the glucose sugar takes the mycelium longer to digest and or break down. Therefore the growth of mycelium liquid culture in the jar may be slower. Thoughts?

@@LoganAddisMusic i did get a good liquid culture going but from there it was difficult to get grain spawn going. I shifted my focus to mostly oysters now and just hunt wild morels. Good luck if you try. I do make a slurry every year and soread it around my property but haven't had any luck with that either.

The pressure pot has so little pressure I imagine altitude may have a significant affect. So what altitude are you at? I’m up where the at pressure is ~75% of what it is at sea level.

Oh the brown color is absolutely sugar decomposing due to Hydration reactions catalyzed by impurities and low ph.

I get yellowing when using malt. Nerve wrecking. Yellow spots (if you get a glob) can also be micellum waste, which is okay.

I’m going to try distilled water . I’ve done many liquid cultures in the past with zero caramelization which is the yellowing color . Anyway I did a few of them in my pressure cooker , the same pressure cooker but they have all yellowed & mycelium will have a hard time eating caramelized sugars . Only thing I did different was using filtered tap instead of using distilled. Maybe my water is really hard or something 🤷‍♂️ no clue but hope switching back to distilled works . Thanks for the video .✌️

did you put for 30 min and the release the valve or let cool and then opned, and same question for grain spawn?

Hey punky!!! Love your vids! When I did all my LC I made it with my PC that reached 11-11.6psi for 40min. I used the same jars, buuuut i would first boil tap water and put it in a QT jar 500 to 600ml, mix it w/ 4% karo (1tbs - under 1 1/2tbs), stir, and pass it to the smaller ones. In this case, more isnt always better, which i did once and it didnt work as well. Also, some folks use Potatoe dextrose w/ karo. Karo alone works fine just for me. I did the 1tbs of karo per jar once as well, and it looked like this and didnt work for me. That's just me tho. All my jars looked really clear.

On the instant pot I did normal heat high pressure, 10 minutes. Put 3 cups of water in, put some jar rings on the bottom. Filled my jars just like yours and put some foil over the tops just to be sure and when I got back and released the pressure the were boiling and over 1/3rd of the liquid in them had completely left the jars. I am very confused by these results. Any advice would be welcome.

Also, microwaving canning jars can cause them to crack in the pressure canner.

I do malt peptone water karo i set it to pressure cook 65 min and it works flawless now it does it cloudy bu5 it settles i put a lil more malt and peptone in my mixture wich i think the clouding is from.once mycellium.start growing it gobbles.up cloud and it clears.up super fas5

you ARE using spore syringes to inoculate the jars. that's spores to LC.

did you try distilled water only without coconut water? also, using raw organic honey instead of karo? you seem to be experimenting and it's helpful!

I'm not sure to understand what is the reason to use distilled water in this case. I understand that you would want clean water to limit possible contamination, especially since you are not using a pressure cooker so the sterilization might not be perfect, but I would think that the minerals brought by the water could be useful to the spore development. And distilled water lakes minerals. Personally I would just go with an unopened bottle of mineral water. On the other hand I would stay away from filtered water, because I think you are referring to water filtered using like a Brita cartridge, those cartridge always have stagnating water in them so they are the perfect place for bacterial development and the activated charcoal will strip most of the minerals.

The coconut water that I add is suppose to add some extra minerals. But honestly my rain water and tap water jars are both doing equally as amazing, so I think the source of water doesnt matter much. :)

I did this the other week for 60 minutes instead of 30 and it caramelized. I’ll try it again on like 15 or 20 minutes. I know the default time on an actual pressure cooker is 15 mins but I’ve had no issues using the boiling method and tap water so maybe if I use distilled it will still work out... WOO! Experiments! Also, I haven’t tested the instant pot on grain jars yet because my rye took over a month to get to me but trying it soon. So I’m not fully recreating your experiment because it’s not millet but it we’ll see. I have some blue oyster mycelium in Karo and it’s thriving. Edit: Throwing this out there, I changed my gasket on my instant pot to one I reserved specifically for sterilization because when you bake something like cheesecake it’s recommended you have another gasket for sweet things otherwise your baked good will taste soupy. I don’t know if it 100% matters because these are jars but just an extra precaution.

The color is due to the things in the water. Distilled water is nothing but pure H20 where filtered, tap etc. has mineral deposits etc that it picks up from the pipes etc. Probably a reaction with the materials in the water.

I believe it's more the caramelization of the sugar, too much heat! You don't need to sterilize lq jars for that long.

the white magnet you put in has only a use if you indeed own a magnetic stiring rod btw, without it it doesn't really do anything, except maybe look nice :P

I think the color is from you overcooking the sugar caramelization, Try less time.

Soak your jar of Lc in boiling water for 3 days, allow to cool. then shake jar for 1 hour str8. Add a dash of liquid soap into insta pot, stare directly at your instapot for about 5-6 minutes give or take. Custom set time for 5-6 hours. Before you know it you're screwed.
