This video is about pyrosequencing method of DNA sequencing. Pyrosequencing is a sequencing technique in which, the incorporation of dNTP in the DNA is detected in The form of light. If dNTP is not incorporatred, no light is detected. hence the sequence information of the DNA can be easily obtained by detecting the light.

Amazing video and great explanatn too but i juz have a silly doubt...can u plzz clarify..if we add a dNTP to the flow cell and LIGHT gets produced, then doesnt it mean dat the corresponding base in the template is complementary to the dNTP added?? For eg. if we add dGTP and get LIGHT, wont the corresponding base in the template be 'C' den?? The video shows dat it wud be 'G'....so im a bit confused..plz clarify sir..thank u in advance..😇

Thank you very much for your question. Yes the corresponding base in the template will be "C". the nucleotide that has been added is "G" The video shows simplified animation about the technique.

There should be an issue here if I understand well. At 3', during the example, the added nucleotides are in the order : G A T C A G A T etc... However when the second template is "GC", dATP is added, and nothing happens for template 2, only template 1 gets light. But then there is dGTP then an other dATP, WHICH THIS TIME CAUSES LIGHT. Why does the first dATP did not cause light in template 2 ? Just a little error on the video or am I missing something ? Great explanation otherwise

How do u know the specific sequence? The dNTPs can be matching the nucleotides in the template strands randomly..

They have to be added in sequence, also, it's the phosphodiester bonding that releases pyrophosphate not hydrogen bonding.

When you look at the flow gram of a pyrosequence, not all the peaks have the same hight. Why is the light sometimes more intense?
