Here is a summary of the key points from the video: - NAD+ is a critical molecule for cellular energy production and repair that declines with age. Boosting NAD+ may promote longevity. - Popular NAD+ precursors like NMN and NR may provide some benefit but don't address the root causes of NAD+ decline. - The root causes are declining NAD+ recycling enzymes and increased inflammation. NMN and NR simply provide more raw material. - Dr. Nichola Conlon created Nuchido TIME+ to target the root causes. It contains nicotinamide plus ingredients to boost NAD+ recycling and reduce inflammation. - A placebo-controlled, double-blind crossover study on TIME+ showed it significantly increased NAD+ levels, NAD+ recycling enzymes, sirtuins, and reduced biological age in 28 days. - Future Nuchido products will likely target cellular senescence. - Dr. Conlon believes future anti-aging treatments will make age-related decline seem inhumane, much like current views on allowing cancer. In summary, the video explains how Nuchido TIME+ targets root causes of NAD+ decline and showed significant anti-aging effects in a robust clinical trial. Future longevity treatments may make aging as we know it obsolete.

Niacin (B3) and niacinimide (nicotinamide) both enter cells through passive transport. No ATP is required. NMN must be converted into NR and then can be absorbed through ACTIVE transport. Niacine and niacinamide are less expensive, can be taken in higher molar concentration, and are likely to be more quickly absorbed. Niacinimide is the primary and limiting storage substrate of the salvage pathway to make NAD. Ribose is present in abundance.

In the trial they measured NAD+ in the blood, because to measure it in the muscle or the brain they would have needed to collect biopsies, which as you can imagine is quite invasive and challenging to do. Glad to hear that is working for you, and of course, we're all free to choose what works best for us 🌱✨ As mentioned in the video, boosting NAD+ through NMN/NR isn't necessarily a bad thing - and indeed, many are benefitting from it. But it's important to remember that boosting NAD+ with just a precursor is a one-sided approach that isn't taking into account the much more complex processes involved. So, that's why there's a price difference. Hope this helps, and best wishes on your longevity journey!

I understand the pathways that lead up to NMN and have tried addressing those myself in the past, but without much success. @catman4471
@catman4471 Год назад
@@CyrilAndPriscilla Hi, I feel that this is just another 'I'm a scientist, buy my product' type of thing. I do understand the reasoning behind it..there are different pathways to creating NAD and the idea behind their supplement is to ensure the de novo pathway is working as it should, rather than relying on the salvage pathway (Niacin, NR, NMN). But all you have to do is get a good nights sleep to create NAD+ through the Kynurenine/de novo pathway. I've noticed that a number of doctors are too engrossed in stats and reports rather than practical experiences of those who take NAD pre-cursors, and I don't think they can really appreciate the benefits we get from NMN. You do not get the same physical or visual benefits by increasing Tryptophan or B6 etc, and you're right, it doesn't make sense according to their approach, to include Niacin.

Follow up NAD+ effects of Time+: Please correct me if math is wrong. NAD+ has a molecular weight of about 660 grams per mole. Typical NAD+ levels appear to be on the order of 40 to 60 micromolar intracellular although some google scholar papers mention blood as opposed to intracellular. The Jinfiniti site which does NAD+ testing reports in micromolar and shows optimal values roughly on the order of 50 micromolar, so expect typical baseline to be lower. Converting 50 micormolar NAD+ to ng/ml I get over 30,000 ng/ml which is huge compared to the increase Nuchido showed of about 17 ng/ml. Calculation below. 660 grams/mole for NAD+ 50 micromolar is 0.00005 mole/L 0.00005 mole/L x 660 gms/mole = 0.033 gm/L 1,000,000,000 ng/gm x 0.033 gm/L = 33,000,000 ng/L 1000 ml/L 33,000,000 ng/L x 1L/1000ml = 33,000 ng/ml If someone here has the skill to do the conversion and check this please do as I don't want to misrepresent Nuchido's results, but so far Nuchido has not clarified what the baseline values of their subjects were or offered up typical values for comparison. One poster noted that he got 33 ng/ml but I did not see his work and he has not yet commented on my details shown above..

Overall, the goal of this episode was to go deeper into the processes around NAD+, while also understanding more of Dr. Nichola's innovative product. It's not that often that supplements are tested with a randomized, double-blinded, placebo-controlled crossover clinical trial, and we believe this says a lot about Dr. Nichola's approach. P.S. If you already take a multivitamin with vitamin C and zinc, then you could just buy nicotinamide, which is about $0.06 per 250 mg , $0.18 per day if you took it 3 times a day.

Why not just take NAD?
@cravarc
@cravarc Год назад
@@HH-gv8mx Great question. NAD is not absorbed well if taken and so you must take a precursor molecule instead. There are three chemical pathways for NAD synthesis: the tryptophan path (not very responsive to supplementation); the niacin path (effective, but getting the dose right is challenging as too much increases inflammation); and, then there's the NR path. NR is turned into NMN so, recently, people have been augmenting NR with NMN or just taking NMN. I've just switched to taking NMN sublingually. The NMN by Renue by Science is what I'm trying. By the way, Nicatonamide is on the same pathway (the Salvage pathway) as NR/NMN, and NMN is also after it in the pathway. If you've heard of people taking Resveratrol or proanthocyanins, too, it's partly to grease the enzymatic wheels for that conversion to NMN. I hope this little ramble expresses well enough my limited understanding.

When is the best time to take this supplement?
@beyondsapiens
@beyondsapiens Год назад
Hi Mike, thanks for your comment! The best option is to take the first 3 pills before your breakfast. While the second dose of 3 capsules should be taken at least 6 hours apart. What I, Alessandro, do, is to take the first dose just a bit before my 1st meal at around 8.30AM, and then last 3 pills with my 3rd meal at around 4PM. Overall, the directions are to take 3 capsules with water twice daily, at least 6 hours apart. It's recommended taking them with food as for some people taking any supplements on an empty stomach can make them nauseous. Depending with how strict you are with your fasting (in case you do it), it should be noted that TIME+ does break the fast. If you take the 1st dose to break your fast, then you extend some benefits, such as AMPK activation. If you need anything, we're always here to help. Thanks and best wishes on your longevity journey 🌱✨

I notice that this product ALSO includes an NAD+ precursor, niacinamide. If the argument is that we don't NEED NMN precursors if we just address the recycling problem, then why bother including an NMN precursor in the Nuchido formula at all? Also, why include the NMN precursor niacinamide and NOT the precursors NMN or NR? Is there some benefit to niacinamide versus NMN or NR?
@beyondsapiens
@beyondsapiens Год назад
Hi Cyril & Priscilla, very good question! The reason why TIME+ uses nicotinamide is because nicotinamide is actually the best precursor in terms of bioavailability, and it's the precursor that our bodies naturally use to make the largest amount of the largest proportion of NAD+. It doesn't require any transporters to get in the cells, and it freely diffuses across membranes. Unlike NR and NMN that do require transport. So, the whole argument is that precursors alone don't address the root cause of the problem, and a whole-system approach is needed to boost NAD+ in the best way. Hope this helps!

@@beyondsapiens Thank you for the comprehensive reply! Can you please refer me to some scientific references that show that nicotinamide is the most bioavailable precursor to NAD? I've read some papers that show that supplementing NMN will raise intercellular NAD, but I have not found any papers that show that supplemental nicotinamide does that. Are there any research papers that, first, show that nicotinamide actually raises intercellular NAD and, second, that it does this BETTER than supplemental NMN? Thanks again!
@CyrilAndPriscilla
@CyrilAndPriscilla Год назад
@@beyondsapiens I did find some papers that show that nicotinamide raises intercellular NAD, so please ignore part of my last question. However, I still haven't found a paper that shows that it's more bioavailable than NMN. For example, while NAM doesn't need a transporter to get into a cell, it is still TWO biochemical steps away from NAD while NMN is only ONE biochemical step away from NAD. That is, it is not at all obvious to me which would produce more NAD in the cell-- NAM or NMN. It is completely possible that that, because it takes an EXTRA STEP to make NMN from NAM that that might trump the fact that it doesn't require a transporter. What I'm saying is, I have found no research that proves that supplemental NAM does a BETTER JOB than supplemental NMN at raising intercellular NAD. I mean, it MIGHT--- but is there actual research on that?

The Nuchido team is excellent at answering any questions you throw at them. I have been taking NMN (1000mg) with TMG and Apigenin for 6 months with great effect. Nuchido has everything in their TIME+ that controls methylation and CD38 levels (although they said you can keep taking TMG if you want, but it's not necessary) so I'm looking forward to starting my TIME+ after finishing my NMN supply in about 7 days time.
@beyondsapiens
@beyondsapiens Год назад
Hi Craig, great question, and thanks for the comment! Generally, TMG is taken alongside NMN and NR to counteract the methyl group depletion that they cause. TMG is not needed alongside Nuchido TIME+ because rather than simply trying to fix falling NAD+ levels by putting more raw material into the cell (using precursors like NR and NMN) it is designed to fix the underlying reasons why NAD+ is actually declining, namely the failing salvage pathway. As the cell can recycle the NAM back into NAD+ it avoids the methylation problem. TMG though, can still provide additional benefits such as improved muscle performance (when combined with high-intensity training) and reduced homocysteine levels. So, you can continue taking it to get those additional benefits :) Hope this helps!

Hi there, thanks for your comment :) To give you more context about the use & quantity of sunflower oil, please see the comment below from the Nuchido team. It is present as an ingredient contained within the Rice Extract Blend we use in Time+, and allows us to avoid the use of other less natural ingredients, such as magnesium stearate, to help the flow of ingredients into the capsule shell. Sunflower oil is present only in a tiny amount. In 6 capsules (daily dose) the sunflower oil provides only 1.7mg linolenic acid (omega 6). As a comparison a 5ml spoon of sunflower oil contains roughly 3250-3500mg linoleic acid (Omega 6). • The sunflower oil is Non GMO verified. In fact, Nuchido Time+ does not use any GMO materials. • The sunflower oil is not heat treated. The oil is expeller pressed. • The sunflower oil is not processed using additives or hexane chemicals. • The sunflower contains no trans fats.

Nicotinamide (NAM) and nicotinamide mononucleotide (NMN) are both precursors of NAD+, but they have different bioavailability. Nicotinamide is a direct precursor of NAD+, but it is less bioavailable than NMN. This is because nicotinamide needs to be converted to NMN by the body before it can be used to produce NAD+. NMN is an indirect precursor of NAD+, but it is more bioavailable than nicotinamide. This is because NMN can be absorbed directly by cells and converted to NAD+ without the need for additional conversion. A study published in 2022 in the Journal of the American Medical Association found that NMN was absorbed more rapidly and efficiently than nicotinamide in humans. The study also found that NMN increased blood NAD+ levels by up to 20%, while nicotinamide had no significant effect. In summary, NMN is the best NAD+ precursor with the highest bioavailability. This means that NMN is more effective at increasing NAD+ levels in the body than nicotinamide. However, it is important to note that more research is needed to determine the long-term efficacy and safety of NMN and nicotinamide. ::::: According to available scientific studies, fisetin is the best natural supplement for activating NAMPT and recycling NAD+. ::::: Fisetin is a flavonoid that is found in fruits such as grapes, strawberries, and onions. In vitro and in vivo studies have shown that fisetin can increase the expression and activity of NAMPT. A study published in 2022 in the Journal of Cellular and Molecular Medicine found that fisetin increased NAMPT expression in human cells by up to 50%. The study also found that fisetin increased NAD+ production, which is an essential cofactor for many cellular functions, including ATP synthesis, DNA repair, and stress response. Another study published in 2022 in the Journal of the American College of Cardiology found that fisetin increased NAMPT activity in mice. The study also found that fisetin reduced inflammation and improved cardiac function in mice. Rutin (quercetin) is another flavonoid that has been associated with NAMPT activation. However, the studies on rutin for NAMPT activation are less conclusive than the studies on fisetin. Resveratrol, EGCG, and coenzyme Q10 are other natural supplements that can activate NAMPT, but they are less effective than fisetin. In summary, fisetin is the best natural supplement for activating NAMPT and recycling NAD+. However, other natural supplements, such as rutin, resveratrol, EGCG, and coenzyme Q10, may also be beneficial. But again Fisitin is better. ------------------------------------------------------------------------------------------- Which cells cause more inflammation? T cells or B cells? In general, T cells are considered to be the main drivers of inflammation. They secrete pro-inflammatory cytokines, such as interleukin-1 (IL-1), interleukin-6 (IL-6), and tumor necrosis factor-alpha (TNF-α), which stimulate the inflammatory response. B cells can also contribute to inflammation, but in a more indirect way. They produce antibodies, which can bind to antigens and trigger the activation of T cells. Additionally, B cells can produce pro-inflammatory cytokines, such as IL-6. However, it is important to note that inflammation is a complex process that involves multiple cell types and molecules. The role of T and B cells in inflammation can vary depending on the type of infection or disease. In some cases, B cells can even suppress inflammation. For example, regulatory B cells (Treg) secrete anti-inflammatory cytokines, such as IL-10. In summary, T cells are considered to be the main drivers of inflammation, but B cells can also contribute to this process. The role of T and B cells in inflammation can vary depending on the type of infection or disease. Basic advice Based on the latest scientific studies, the best way to reduce inflammation is to take Fisetin and NMN and you can also take Apigenin (optional) in the form of powdered parsley. Fisetin will lower CD38 inflammation in T cells by up to 70% and apigenin CD38 in B cells by up to 80%. Plus, fisetin will activate NAMPT and still eliminate senescent cells.

Hi Daniel, thanks for your input! We want to provide a comprehensive reply, so please find below the reply directly from the Nuchido team. We hope it helps! "NAM is our chosen precursor in Nuchido TIME+ because it has increased bioavailability over NMN. NAM is an uncharged molecule that can rapidly diffuse across the membrane of any cell and mitochondrial membranes. On the other hand, NR and NMN depend on specific transporters to allow them to cross the cell membrane (ENTs and SLC12A8 respectively). This means bioavailability of these precursors is dependent on whether the cell type has the transporter or not (and many do not). Additionally, with NMN some studies have suggested that the majority of NMN is metabolised into NR before it is taken up by the cells. NAM is also the bodies preferred precursors as it is the breakdown product of NAD+ consumption. Nuchido TIME+ increases activity of NAMPT which is the enzyme that converts NAM to NMN, therefore this conversion process does not limit the production of NAD+. As you mention there are very promising studies with fisetin, however it has limited bioavailability in humans. Further human clinical trials are needed to consolidate the pre-clinical benefits of fisetin and work is needed to improve its bioavailability. Nuchido TIME+ contains saphora japonica because it contains rutin a glycoside of quercetin, this greatly increases the bioavailability of the quercetin in the body. We also know from our human clinical trials that Nuchido significantly increases the activity of NAMPT to restore NAD+ recycling. When designing the formulation for Nuchido TIME+ we used a systems Pharmacology approach to identify active ingredients against specific targets. Each ingredient was included and formulated to ensure maximum bioavailability and our approach is supported by evidence from our human clinical trials. Our lead scientist Dr Conlon has published several papers about this -> Conlon, N., & Ford, D. (2022). A systems-approach to NAD+ restoration. Biochemical pharmacology, 198, 114946."

According to the latest studies from Google Scholar, both NAD+ precursors NR (nicotinamide riboside) and NAM (nicotinamide) have beneficial effects against aging and aging-related diseases in animal models. However, some studies suggest that NR may be superior to NAM in terms of efficacy and safety. For example, one study compared the effects of NR and NAM in mice with Alzheimer's disease and showed that NR significantly increased NAD+ levels in the brain and blood, while NAM had no effect. Moreover, NR improved cognitive function, reduced inflammation, and protected mitochondria in mice, while NAM had no such benefits. Another study compared the effects of NR and NAM in healthy humans and showed that NR was well tolerated and increased NAD+ levels in the blood by up to 2.7 times, while NAM caused side effects such as nausea, fatigue, and headache and did not increase NAD+ levels. Therefore, based on this evidence, NR seems to be a more effective and safe NAD+ precursor than NAM. However, more studies are needed to confirm these findings and explore the molecular mechanisms involved. I have now been taking Nuchido Time+ for 8 weeks at the dose you recommended (2x3 capsules daily with meals, with at least 6 hours between each dose). It makes no sense to take an additional NMN product.. I would like to take you on a short journey to see how the product works for me: - After 7 days: I have a deeper sleep. Although I still wake up 1-2 times during the night, I am much better rested in the morning! - After 14 days: In addition, I no longer have chronically cold hands and have not been so sensitive to cold for a long time before taking Nuchido Time+. Maybe Nuchido Time+ even has a positive influence on the blood circulation! Best regards from Germany and thank you for the really great product. I am curious to see what effect the consistent use of the product will have on me. Before I started taking Nuchido Time+, I had my biological age tested via GlycanAge. I will have the results in 2-3 weeks. After one year of taking the product, I will repeat the test and then compare how my biological age has developed. A very exciting story 😊 But if you look into the science behind the product, you can certainly hope for further very positive effects, here I am thinking especially of the preventive prevention of serious diseases. The system pharmacological approach of Nuchido convinces me very much. - After 21 days: I have been doing fitness sports for many years and therefore probably have a relatively high NAD level in relation to my age. So far, I have not been able to directly feel an increase in energy. But now I have done a test. So far, and for years, I have not managed to do more than 154 pull-ups in 5 sets. I usually managed between 148 and 152 pull-ups. Now I did the test after 3 weeks and I am positively surprised. I now managed to do 165 repetitions in 5 pull-up sets. This might not sound like a big increase (between 7-11%), but for me personally this is a very positive surprise after years of stagnation. Since I haven't changed any other dietary or training habits, the increase in strength endurance can be chalked up to Nuchido Time+. Overall, I recover faster between sets and I am also ready to go again faster after the training sessions. On 23.09.2023 (i.e. before taking Nuchido Time+) I had my biological age tested by GlycanAge for the first time. On 09.10.2023 I received my result. I would have to lie if I said I wasn't a bit excited.  My expectation was already tending towards the positive, which means I was already expecting my biological age to be slightly lower than my chronological age. However, the actual result surprised me very positively. My biological age is 23 years lower than my chronological age. It felt like a birthday ;-) But the result is not set in stone and motivates me to continue to lead as healthy a lifestyle as possible, to make sure I get enough exercise, fitness training and sleep. I will also continue to take the dietary supplements I have been taking so far, such as curcumin, astaxanthin, resveratol, etc. I am very excited about the results. For me, it will be very exciting when I do the test again in just under a year's time to see if I can go one step further with Nuchido Time+. In any case, my anecdotal experiences in the first 3 weeks of taking it are extremely positive. - After 28 days: A further increase in strength endurance has been recorded. In addition to 170 pull-ups (back) in 5 sets, increases of between 3-10% have also been recorded in all other muscle groups. For the first time in the 4th week of taking Nuchido Time+, an improved metabolism is also noticeable, which enables me to maintain my weight more easily. The ratio of muscle to fat should therefore also improve. All other already existing positive effects of the first weeks continue undiminished. - After 5 weeks: Visible reduction in belly and hip fat. Overall, my skin is becoming softer and the first very slight improvements in the appearance of my skin can be seen. In the meantime, I have increased the number of pull-ups to an incredible 180 repetitions in 5 sets. All other exercises have also improved by between 5-20% since I started taking Nuchido Time +! The muscle pain after the training sessions is reduced and also gone much faster. More by accident, I have discovered that I have become faster when walking. Overall, I feel an energy boost that I had not thought possible. The already mentioned benefits from the first 4 weeks all remain (deeper sleep, reduced sensitivity to cold, longer endurance, faster regeneration, improved metabolism, improved weight control). - Continued after 6 weeks of use: All the previous benefits of the first 5 weeks are retained, and in some cases even enhanced. The complexion is noticeably better, finer-pored and fresher. I was able to increase the number of pull-ups in 5 sets to 187! There has also been steady progress with other exercises (legs, shoulders, etc.). - Continuation of my experience with Nuchido Time+ after 7 weeks: There are still slight performance boosts in terms of strength - and endurance. This applies to all muscle groups. I have now completed 192 pull-ups in 5 sets. That's about 28% more than I was able to do before taking Nuchido Time+. In the meantime, I can also feel and see that my skin is becoming more elastic. Apart from that, all the positive experiences of the past weeks remain. It's remarkable how much energy Nuchido Time+ gives me after just 7 weeks. I am curious to see whether there will be further improvements. Perhaps my NAD+ level is now slowly being topped up to such an extent that in future even more resources can be used for secondary tasks in the cells, such as for skin and hair. So far, I am completely thrilled. - Ongoing experience with Nuchido Time+ after 8 weeks: Further improvements in the appearance of the skin are recognisable, smaller wrinkles are slowly disappearing. There has already been an improvement in the overall contours of my face. I wouldn't have thought it possible, but my hair is slowly starting to darken (white hair is turning light grey and light grey is turning dark grey). I have no idea whether this will continue and whether I will even get my natural dark brown hair colour back. However, I am realising one thing more and more. Among other things, Nuchido Time+ improves my metabolism considerably. Nutrients that I take with my meals and food supplements that I have been taking for years (curcumin, astaxanthin, resveratol, OPC, hyaluronic acid capsules) seem to have considerable positive synergistic effects when I also take Nuchido Time+! I am convinced that the food supplements I have already been taking can be processed much better in the cells. For some time now, I have been following Dr Nichola Conlon's numerous and very informative presentations very closely. Among other things, I learnt that the body cannot distinguish the source of the nutrients it receives (food, food supplements or medication). Incidentally, my strength is still increasing, for example I can now do 197 pull-ups in 5 sets. Am I enthusiastic about Nuchido Time+? Yes, I am 😇

Seems unusual to hear a doctor with such lazy pronunciation constantly dropping Ts’ ie longevi’ee, scien’ists, wha’ , thoh’ of, lod of, me’abolic, star’alin. Also developin’ poot in’ Also terms like “kind of like” “grab on to i’ “ the over and in appropriate use of the word “like” and “yah know” I just can’t imagine that language in a PHD thesis or a scientific presentation ti piers” Sounds very much like an advert complete with smiley faces of appropriately aged actors. Am waiting for the bit encouraging to buy now before those nasty big pharma companies take the add down! Maybe I am being unfair but it is unusual. Doesn’t instil confidence!

There is no such thing as longevity. I dont know why this word gets thrown sround do much. You start sging st 40, snd are old by 50. Sging is hard, brutal and ehy would a person want to live in a decaying body for 100 yrsrs?

Really ? What poor outlook you have ! I know many people in their 80s that still work and are very active all day. I'm 59 and more athletic than most 30 year olds.

First of all you start aging the minute you're born not at 40 and I don't know in whose book your old at 50 except yours. And as long as I'm not in a diaper and no one has to feed me I want to live to be 100

@@christinamerklin2166 , You do understand you will be decaying beyond belief till 100? That's no quality of life.

Nasty arthritis, decaying of joints, nasty skin aging, arthritis. It's all very horrible to look at. Nobody in their right mind would think aging is good. You'd have to be very naive to think loss of mobility, stamina, and just feeling alive loss is good. If you didn't, you wouldn't be on longevity sights. There is no longevity, lol. It's just decaying, and die by 70. It's terrible
