As someone who has already performed multiple TLC experiments, I feel that this video will definitely help students who want to watch how to perform this experiment before actually doing it in class. This has the upside of the student focusing on the understanding part instead of focusing on how to simply perform the experiment. I also appreciate that this youtube channel highly recommends watching an in-depth video of TLC before watching how to perform it in lab.

In addition, if you wish to reuse a capillary you can dip it in acetone to clean it between spottings

TLC can be used quantatively by using different concentrations of the same standard, and comparing the sample spot size to the standards.

Excellent, but you should make many examples for differnt sustances,polar or non polar,in addtion you can explain column chromat. and HPLC.

I have a qustion. What if the extract sample we have is semi-solid, how do we dissolve it? and how to choose the appropriate solvent for a sample that has never been studied?

great video, hope you do one one column chromatography (adsorption or flash chromatography) and explain how tlc can be used to effectively create an efficient solvent system for any purification work-up using the more common column chromatography. Can a standard UV-C light (~253nm, for disinfecting) be used for visualizing plates at home? they are only like $10, compared to the mineral lights or light boxes used in labs which are hundreds of dollars. when creating reference samples for the tlc spotting of known or unknown samples, do they have to be in aqueous solution or in a solvent, and does it matter how dilute the sample is? How do you create an Iodine chamber for developing tlc plates?

What's the purpose of capping the solvent and letting it sit, before introducing the plate?

Letting the tlc tank become saturated with the solvent vapour means that the solvent doesn’t evaporate from the leading edge as it rises up. This slows down the rate of movement and the components of the mixture separate into better defined spots rather than streaks. There are some times when this isn’t needed, so you can run the separation without tank equilibration. Also, many of the solvent mixes used are not “nice” chemicals to have in the air…
