SIRT6 & FUCOIDAN: In today's video, we examine the new paper that just came out on activating SIRT6 and the supplement extracted from seaweed that activates it.
We'll do a quick review of what sirtuins are and how they function before jumping into the new paper that was published last month on SIRT6. Prof. Haim Cohen and his team extended the lifespan of both male and female mice by as much as 27% (or over 30%, depending on who you believe). OK, I looked into this further. Fucoidan is not the name of a specific chemical substance, but rather the name for a class of substances. There are several different types of fucoidan, each with different molecular structure and different properties. Fucoidan refers to a type of polysaccharide that contains substantial percentages of L-fucose and sulfate ester group, but can also contain other monsaccharides, including mannose, galactose, glucose, xylose, etc. plus other compounds like uronic acids, acetyl groups and proteins. What type of fuciodan a compound is can depend on the type of seaweed that is was extracted from and the method of extraction, to name a few. In light of this, it's not too difficult to see why it can be so hard to find fucoidan that can activate SIRT6. Hope this helps.

Thanks for the info. I watched this video when it first came out. And I've been curious about the Do Not Age product. But I haven't tried it. I keep hoping to hear more info about SIRT 6 activators, specifically in humans.

Great video, thank you. I just ordered the Sirt6 Activator. I currently take Pure Apigenin, Fisetin and CA-AKG. I also take Metformin, even though I do not have Diabetes. I'm 2 months from my 68th birthday. I'm in great health and I have lots of energy. I go to the gym 5 days a week and do weights and aerobics exercising for about 2 hours each day. I love life. Live long enough to live forever!

@@peterdavila3045 I do sympathise as I'm in the same boat. I'm on a low income myself and struggling to afford these longevity supplements, but personally I think NMN is one of the most important. I suppose exercising will help boost NAD levels and maybe taking some dried parsley to keep your cd38 levels down could also help.

Not sure if metformin is really so great. I don't remember where exactly I saw that but apparently the effect on healthy subjects longevity is null and the study showing metformin increases longevity on diabetic subjects has some flaws. Metformin could also be detrimental: each drug has its side effects. And metformin should be avoided on days you have a workout, 5 days a week in your case. I you assume metformin could help by reducing blood glucose (by limiting glucose production in the liver) maybe you could replace metformin by a low-carb diet and/or a 1 or 2 days fast every now and then.

CONGRATULATIONS Lance! I think your decision to create a longevity course and charge for it in order to support your work on RU-vid is brilliant. Your current videos bring all the detail of the complex chemistry and biology of longevity down to a level of understanding better than anyone else on RU-vid. I know it takes a lot of time and work to do that, and I’m glad to support you in your ongoing efforts. This will, no doubt, be a win-win for everyone. You get support and your audience gets a pre-digested source of understanding and information for the ongoing longevity research and the resulting practical capabilities being produced in this complex field. Here are two aspects of the course that I personally would like to see. First, I would really like to have the course content available in some “printed” form in addition to or instead of video. The amount of information involved and its complexity are very large, and just having a video to watch in order for me to mentally capture everything is not adequate. If only video content were presented, I’d have to spend a large amount of time taking notes and creating complex diagrams that I could use for study and review to ensure that I can confidently reason about what I should be doing to extend my own longevity. Ideally, the course would be presented both in video and in a printed format, such as PDF. If that were done, we students could view the video first to get the big ideas involved, and then study the text and diagrams to cement the details in our heads. Second, a hybrid of one-time purchase and a subscription model of pricing might work well. A person could buy the course to get the first version. The course would have to be updated occasionally, maybe on a quarterly basis, in order to keep up with the rapidly evolving field. So a person could either re-buy the course at full price whenever they think it’s worth doing, or they could subscribe to updates after they have purchased the full course so they could have lower cost access to the updates on a continuous basis. And if you really want to confuse everyone, you could also create two tiers of the course - one lower cost tier of video-only, and a higher cost tier of video and printed material. I’m not trying to tell you how to run your course, Lance; I’m just casting my vote for course features I’d like to see. However you do it, I’m sure it will be a major contribution to the many people who don’t have PhDs in this field but still want to understand it in order to extend their lifespan and healthspan. Thanks so much for doing this!

Thanks, Frank. You have no idea how gratifying it is to hear your type of comment. I really wasn't sure how well such a course would be accepted by this community. FYI, I'm planning on including in the course a number of PDFs that would contain additional information. For example, rather than list all of the supplements that I take in the video, which would be boring to listen to, I'd list them in a PDF. I'd also provide PDFs of any charts or diagrams that I use in the video. But I could also include a PDF of the script, perhaps broken down into bullet points for easier reference. With regard to your second tip, I was planning on making the course be available to anyone who signed up for it for life, including regular updates as more information becomes available. But I might like your idea better. I'm really looking for feedback from this community, I want to course to be what they want. I'll probably offer the first edition of the course at a discounted price, on the condition that everyone give me feedback regarding what else they'd like to see in the course, and then that revised edition will be version 1.0. I started taking SupaFuco fucoidan chewable tablets from Tasmania and this has also increased my energy and appetite. If it’s a fucoidan that doesn’t activate my Sirt6, I have no idea, but it’s given me a lot of energy and costs way less than the DoNotAge product, which I can’t afford.

No, there's not. But it makes sense to me. However, depending on your age, there are different strategies to use to make sure that you NAD+ levels are on target. Great video lance. I've watched some of the other peoples videos and I was confused as well. And it seems like there is still some confusion remaining. I'm suspecting the researchers are now influenced by $$$ and might be making some less conservative claims. Thanks for sorting through all of this and boiling it down to 20mins. It must have taken you a long time! I'm going to wait a few more months for more coherent results. One question. If NMN activates all Sirtuins, why bother with Fucoidan? Or is it that NMN only activates the de-acetilysation part of SIRT6?

There are many molecules considered giving longevity. Like taurine, metformine, glysine, nac, creatine, resveratrol,nmn, ca akg etc. All these can be swallowed together or should maintain a distance minimum time between each molecule

You have to take her word that she has a special kind of fuciodan, even though she doesn't explain why her fuciodan is different from the fuciodan you can buy by the kilogram.

Hello Lance, I seem to be the only person using cyanidin as a SIRT6 activator-via ribosylation, as well. Have been taking 200mg daily for two months. Fucoidan can occasionally work at 'cross-purposes' methinks, yet cyanidin is potent and rarely an inhibitor. Thoughts please.

Interesting. Apparently, cyanidin DOES activate SIRT6, and it apparently does it better than fucoidan. I think I'll start taking cyanidin, so that makes two of us. 😀

Lance, Good to hear from you. So now we are 'two' taking cyanidin. . . . Before you commence, be advised of a possible reason why it hasn't yet been commercialized: cyanidin is one of the most aggressively staining substances I've encountered, which could possibly alarm some folks. If you would like more information on titration/dosages, perhaps we will communicate privately.

I've been taking it for eight months. My Twitter page deals with my SIRT6 use and more, but the cyanidin does seem to be working, even though saying that something is working is tricky in the absence of any health issues at age 70. As for an immediate sensation associated with it, none. Cyanidin is incredibly staining and have had a couple of messes to clear up. It literally stains stainless steel so can see why selling it retail might be problematic. If I didn't know it won't irrevocably turn your body purple, I’d be afraid to take it. One other thing: the activator fucoidan can also inhibit SIRT6 when taken at higher dosages, but I understand this is not an issue with cyanidin. Let me know. . . .

Have you been taking Metformin for diabetes or anti-aging/both? 20 years is a long time…. I only have access to Berberine at this point….this is so interesting, about cyanidin, I have no concerns about staining whatsoever.I wonder if you did epigenetic age test or blood marker one. Appreciate all the information that you can share!

Anthocyandin(iirc) is the best SIRT6 activator by like 13x. Found in dark berries, elderberries have highest concentration. Even at 13x the next best, it is tiny on the grand scale. Some might say a fools errand.

Hi Lance, I like your presentations. Please add to your information presented which (natural food) sources provide TMG. For example there is no reason for people like me who eat a steady amount of Wheat Germ, Beets, Spinach, Quinoa, to fork over $100 for TMG supplements, which I would venture to hypothesize are not nearly as bio available and surely not as broadly nutritious as the natural sources as follows: Here’s the amount of TMG found in a 3.5-ounce (100-gram) serving of several foods (24Trusted Source, 25Trusted Source, 26Trusted Source): Wheat bran: 1,339 mg Wheat germ: 1,241 mg Spinach: 600-645 mg Quinoa: 390 mg Beets: 114-297 mg Shrimp: 219 mg Wheat bread: 201 mg Keep in mind that several factors impact the amount of TMG found in foods. For example, certain cooking methods, particularly boiling, significantly decrease the TMG content .

I’m not sure where you’re seeing TMG costing so much, but Prohealth has 100g of TMG powder for under $14 after tax. That’s 14 cents a day for 1g of pure TMG. You’ll pay way more than that from food sources. You won’t get 1g of TMG for 14 cents anywhere else

So what they're selling for that price is Fucus Vesiculosus. Can't you just take bladderwrack which is extremely high in it? On top of that, if you combine the bladderwrack with sea moss and burdock root and you get all of the minerals the body needs? Dr. Sebi used to talk about Fucus decades ago for extending longevity.

I think the Do Not Age Sirt6 activator may be a total scam. Somebody needs to do a third party test to see if it activates Sirt6. When the people making it and selling it are the same ones testing it there's a potential for fraud. There needs to be third party testing for this product and so far as I know there isn't any.

Good point. The supplement is insanely expensive and there is zero human evidence that it will activate SIRT-6 to the extent claimed for it. All studies have involved rodents and much of what works for rodents doesn't produce the same effects in humans.I think any sensible and intelligent person would not be lured by the claims made about this expensive product without first viewing some human evidence to show it works as advertised. The scientist pushing this product is paid by the company that sells it, so is not exactly an unbiased source!

Agreed. Especially when it is going to cost someone over$4,000 a year. Part of the high cost is due to the constant testing of each batch to make sure it works.

I agree as well, as any sensible person would. There is absolutely no proof it works in humans and that fishy “selection” process…they test it how exactly? Do Not Age CEO talking about it in interview - he sound so uneducated, he has no idea what he is selling but def enjoys the revenue I reckon.

Leading edge products done by leading edge teams must start somewhere with the categorization of seaweeds know to be beneficial. There is little profit in vitamins, minerals, botanicals until they are synthesized and commoditized or made into a pharmaceutical. A large number of supplements for sale are ineffective.

Do Not Age sends a sample of every batch of their products out for purity testing. If the molecule they are selling is the same molecule used in the studies are the same, then where is the scam? If Do Not Age was a scam, they would have a lot more products available, like blends and multivitamins, etc.

I bought the starter package including SIRT6 from the this company 8 or 9 months ago and I didn’t notice any positive difference. I get headaches and itching every time I take it and it has a fish smell. Did I receive a bad patch? Did anyone experience something similar?

I've been using SIRT6 Activator for about 6 months and I've noticed no fishy smell nor do I get headaches or itching. But that's just me, everyone is different. But my batches of the product do not smell fishy. I'd contact DoNotAge and let them know. I've found them to be very responsive.

Isn't there a way where you can accomplish the same thing at home through exercise and diet or some kind of lifestyle intervention instead of expensive nostrums which as some of the comments indicate, have the potential for severe gastrointestinal distress and who knows what else?

The seaweed extracts in question cost almost nothing and are not patentable: in just a few months, the competing companies will have had the time to formulate exactly the same fucoidan extract for much less: which is why they want you are subscribing now for a year, at shockingly expensive prices, because they are the only ones on the market, but they know it can't last long... Nonetheless I really consider their brand is a very good one, especially for thieur apigenin at a very competitive price... Anyway, Thanks and thumb up, Lance, for the excellent review !

I agree, DoNotAge is a great brand, which is why I doubt they're trying to scam us with their fucoidan supplement. Fucoidan isn't just one type of seaweed, it's a family of seaweed and as I understand it, not all fucoidan seaweeds produce the sirtuin activator. Which is why they have to test many different samples, which is why it's so expensive. That's my take anyway.

There is a clear conflict of interest because ppl “testing” it and “selling” are the same. And their testing methods never disclosed. This is just so shady, reading description section for that product on their website just made me cringe.

median lifespan is how long most of the mice live. Max lifespan is how long the oldest mouse lives. If most mice live 48 months, then 30% increase is (48 x .3). If Max is 58 months then max is 58 x it's percentage increase. Thus max lifespan could still be higher than median lifespan notwithstanding the fact that median increased by a greater percentage value.

@@LanceHitchings the median lifespan was extended 27 percent, the male max lifespan was extended 11 percent. If the median was 48 months, then it increased by about 14 to 15 months. If the max male was 58 months then it increased by about 6 months. The max male is still older than the median.

I've read that the most powerful sirt6 activator of all is a substance called cyanidin found in such things as the brightly-colored skins of berries like elderberry, blackberry, cranberry, blueberry. Being pretty ignorant about this area, I don't know if those sources of cyanidin would affect the deacetylation or the ribosylation function. May I please ask you for your opinion on the possible sirt6 benefits of taking extracts of these natural flavonoids above and whether they might have the same benefits as fucoidan if taken in large enough quantities?

I took two capsules of Do Not Age Sirt 6 Activator and afterwards was taken to rhe hospital emergency. Got terrible stomach cramps that needed to treated with an infusion. They asked me what I had taken, I told them about the product. They never heard of it - of course. I am sure the cramps came.from the capsules. I never had pain like that before in my whole life. I won't take any of these capsules ever again. Even though, they were damn expensive.

@@james575730 Normally I'm not allergic to seaweed. I eat all kinds of seaweed regularly. Nothing happens. No I did not tell DoNotAge about it yet. Not sure whether I should do it. I consider it as an experiment. So the product might work quite well for many people, and I was in the unlucky posution that my body did not like it.
